Study of the bronchodilating effect of three doses of nebulized oxitropium bromide in asthmatic preschool children using the forced oscillation technique

Abstract: Oxitropium bromide is a potent and long-acting bronchodilator in preschool children at a dose of 750 micrograms and 1500 micrograms. No side-effects were observed. The exact duration of action remains uncertain, but even 4 h after inhaling 750 or 1500 micrograms of OB no additive bronchodilation induced by fenoterol could be observed.

“…In placebo-controlled studies, FOT was useful to document therapeutic response to bronchodilators, such as theophylline, terbutaline, or ipratropium bromide (197)(198)(199)(200)(201)(202)(203)(204)(205)(206)(207). The criteria (and cutoff values) for diagnosis of meaningful reversibility postbronchodilation are reported in Table 9.…”

“…In young children, FOT has been shown to provide a useful and objective method to assess airway responses to bronchodilator drugs, such as metaproterenol, ipratropium bromide or salbutamol versus placebo [87][88][89][90], and to characterise dose/response curves [88,91]. For example, the bronchodilating effect of nebulised oxitropium bromide (750 and 1500 mg) in preschool asthmatic children was shown to last for up to 4 h postinhalation, whereas no additive bronchodilation by fenoterol could be shown [92] .…”

“…The measured resistance of the respiratory system (Rrs) is most closely linked to airway resistance. FOT has been used in clinical trials to document response to bronchodilator inhalation (14,15) and anti-inflammatory administration (16) and has been found to be reproducible and to correlate with FEV 1 . (17) Unfortunately, in the CF preschool population, results have been variable in the ability to detect early disease.…”
