Study of method and system for diagnosis of cancer using autofluorescence and Raman spectroscopy

Abstract: induced fluorescence(LIF) and Raman spectra were measured from normal and tumorous human serum in an attempt to discover some values useful in discrimination between normal and tumorous cases. Red shift of fluorescence peak and decrease of fluorescence intensity were observed after samples radiated by laser. According to one thousand twenty-two samples' spectra, three parameters α, β and δγ are introduced to distinguish normal, benign and malignant from one another. The application of such … Show more

“…In this study, we developed a novel method by which to simultaneously measure the serum autofluorescence and cfDNA-related fluorescence using a real-time PCR system as a convenient fluorometer with temperature and time control “functions”. Compared with the conventional method for detecting serum autofluorescence, which relies on the use of a spectrofluorometer [ 10 12 ], our method is rapid, high-throughput and efficient. All operations could be completed within half an hour.…”

“…The serum autofluorescence intensity reportedly gradually increased as liver fibrosis progressed in rats [ 24 ]. Changes in the serum autofluorescence have been exploited for the diagnosis of chronic liver diseases [ 10 12 ]. Additionally, It was reported that indirect bilirubin was more fluorescent than direct bilirubin in bile [ 25 ], but we did not found this phenomenon in serum as the correlation coefficients of indirect bilirubin were similar with direct bilirubin ( Table S2 ).…”

“…This effect may be partly compensated (approximately by 5% to 10%) in a multimodal approach, 21 when each modality uses optical parameters associated with different tissue components altering during the cancerous process. The combination of RS and AF has been demonstrated for gastric, 21 lung, 22,23 liver, 24 and breast tissues model 25 experiments. Yet the exact RS-AF diagnostic accuracy is still unclear for different skin tumor types.…”

“…Serum-based detection is simple, practical, easily repeated and minimally invasive. Serum autofluorescence is a potential biomarker for liver diseases [ 10 ], with an accuracy of 85% for diagnosing liver cancer [ 11 ]. Cell-free circulating DNA (cfDNA) in serum has been reported as a new tumor marker based on increases in various malignancies, including HCC [ 12 ].…”
