Distribution of lymphocyte subpopulations along the small intestine lymphoid tissue has been examined in 1-month-old lambs using flow cytometric and immunohistochemical techniques. Monoclonal antibodies against CD4, CD8, gamma delta, CD45R and B receptors have been employed in samples from continuous ileal Peyer's patch (IPP), discrete jejunal Peyer's patches (JPP), ileocaecal valve lymphoid tissue (ICVPP), mesenteric lymph node (MLN) and intra-epithelial (IEL) and lamina propria (LPL) lymphocytes. Histological studies were also done. Differences in the lymphocyte distribution have been observed between some of the regions examined, especially between IPP and JPP for most of the markers. A remarkable feature was the existence of morphological and lymphocyte distribution differences between ICVPP and IPP, locations that had been traditionally considered as similar. The antibody against CD45R receptor used in this study, that was supposed to mark B cells and some T cells, detected cell populations located in the dome of the follicles in all the samples, whereas the centre was negative. Lymphocytes positive to the B marker employed were located mainly in the centre, suggesting that both antibodies would mark B cells in different maturation status.

Abstract. Porcine enzootic pneumonia (PEN), caused by Mycoplasma hyopneumoniae (Mh), has been described in pigs in all geographic areas. The disease is characterized by high morbidity and low mortality rates in intensive swine production systems. A morphologic and immunohistochemical study was done to determine the cellular populations present in lung parenchyma of infected pigs, with special attention to the bronchusassociated lymphoid tissue (BALT). Polyclonal and monoclonal antibodies were used for the detection of antigens of Mh, T lymphocytes (CD3 ϩ , CD4 ϩ , and CD8 ϩ ), IgG ϩ or IgA ϩ lymphocytes, and cells containing lysozyme, S-100 protein, major histocompatibility complex class II antigen or myeloid-histiocyte antigen. Findings in lung tissues associated with Mh infection were catarrhal bronchointerstitial pneumonia, with infiltration of inflammatory cells in the lamina propria of bronchi and bronchioles and alveolar septa. Hyperplasia of mononuclear cells in the BALT areas was the most significant histologic change. The BALT showed a high morphologic and cellular organization. Macrophages and B lymphocytes were the main cellular components of germinal centers. T lymphocytes were primarily located in perifollicular areas of the BALT, lamina propria and within the airway epithelium, and plasma cells containing IgG or IgA at the periphery of the BALT, in the lamina propria of bronchi and bronchioles, in alveolar septa, and around bronchial submucosal glands. The hyperplastic BALT in PEN cases consisted of macrophages, dendritic cells, T and B lymphocytes, and IgG ϩ and IgA ϩ plasma cells. CD4 ϩ cells predominated over CD8 ϩ cells. Local humoral immunity appears to play an important role in the infection.

Small ruminants, like goats, would make excellent animal models for not only infectious diseases in large ruminants but also analogous diseases in humans, such as human tuberculosis, Crohn's disease, melioidosis and brucellosis. The main disadvantage for the small ruminant model is the lack of sufficient baseline data on normal, healthy goat kids. Furthermore, most reagents (antibodies and the like) were not developed for goats or sheep. It is important to demonstrate that available resources, especially from the bovine system, cross-react with the caprine and/or ovine system. Finally, potential breed differences have to be evaluated before goat or sheep studies are compared. In this study, leukocyte cell populations were defined in twenty-six dairy goat kids via flow cytometry. We report no significant differences between three breeds of dairy goat kids and demonstrate the effective use of various antibodies for caprine immune cell markers. No breed-specific differences were detected for any leukocyte cell population or for markers specific for various antigen-presenting cells or T cell populations. Interestingly, however, statistical significant differences were found for leukocyte cell populations for the two different time points two weeks apart presented in this study.
