Study of Kidney Repair Mechanism of Cassava Leaf Extract on Gentamicin induced Nephrotoxicity

Abstract

AbstrakKonsumsi obat obatan seperti gentamisin dapat menyebabkan kerusakan dan penurunan fungsi ginjal. Daun singkong banyak mengandung mikronutrien, seperti vitamin C, vitamin A, dan karotenoid, yang dapat berfungsi sebagai antioksidan. Hasil penelitian yang pernah dilakukan oleh Almashuri membuktikan bahwa daun singkong mempunyai kadar senyawa karotenoid yang tinggi. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak daun singkong terhadap perbaikan fibrosis ginjal mencit secara invivo. Penelitian ini merupakan true experimental dimana menggunakan hewan coba mencit (mus musculus) yang dikelompokkan menjadi 5 kelompok secara acak. Kelompok-kelompok tersebut adalah 2 kelompok kontrol (positif dan negatif) dan 3 kelompok perlakuan (daun singkong 150, 300, dan 450 mg/hari). Pada kelompok kontrol positif dan perlakuan mencit dibuat model fibrosis ginjal dengan cara memberikan gentamisin 80 mg/kgBB/hari. Perlakuan dilakukan setiap hari selama 14 hari kemudian diamati kadar serum kreatinin beserta skor kerusakan histopatologi ginjalnya. Analisis statistik menunjukkan adanya penurunan bermakna kadar serum kreatinin (p<0.05) dan penurunan bermakna skor histopatologis ginjal. Hal ini menunjukkan bahwa ekstrak daun singkong dapat memperbaiki kerusakan ginjal akibat fibrosis ginjal pada mencit. Kata kunci : ekstrak daun singkong, kreatinin, histopatologi ginjal Abstract Drugs consumption like gentamicin caused damage and decreased of kidney function. Cassava leaf contain lots of micronutrients, such as vitamin C, vitamin A and carotenoids, which also as an antioxidant. The results of the study conducted by Almashuri proved that cassava leaf have high levels of carotenoid compounds. The purpose of this study was to determine the effect of cassava leaf extract on the repairment of renal damage in mice invivo. This research is true experimental using mice (mus musculus) and were grouped into 5 groups. Two control groups (positive and negative) and 3 treatment groups (cassava leaves 150, 300, and 450 mg / day). Gentamicin is given every day for 14 days and then observed on serum levels of creatinine along with histopathological scores of kidneys' damage. Statistical analysis showed a significant reduction in serum levels of creatinine (P <0.05) and a significant decrease in renal histopathologic score. This study suggests that cassava leaf extract can repair kidney damage as a result gentamicin-induced nephrotoxicity in mice.
