Study of intra-varietal diversity in biotypes of Aglianico and Muscat of Alexandria (Vitis viniferaL.) cultivars

Abstract: Background and Aims: Somatic mutations are the major source of intra-varietal variability in some species such as grapevine (Vitis vinifera L.) that are propagated vegetatively. Aglianico and Muscat of Alexandria are two cultivars mostly cultivated in the south of Italy, showing great intra-varietal diversity. The variability of 15 clones of Aglianico belonging to three biotypes, Taburno, Taurasi and Vulture, and 21 clones of Muscat of Alexandria, coming from France, Greece, Italy and Spain, was investigated. … Show more

“…The carotenoid and chlorophyll content of the berry samples was determined by high‐performance liquid chromatography (HPLC) as previously described (Mendes‐Pinto et al ., ) with minor modifications (Kamffer et al ., ). The norisoprenoid content was determined during ripening by solid‐phase micro‐extraction and gas chromatography/mass spectrometry (GC‐MS) as previously described (De Lorenzis et al ., ). The flavonol and HCA content was determined by HPLC as previously described (De Lorenzis et al ., ).…”

The flavonol and HCA content was determined by HPLC as previously described (De Lorenzis et al ., ). In each case, 50 berries were used for extraction.…”

“…In this project, a total of 18,071 SNPs were selected, a third of which (4510 SNPs) identified in the Northern American species genome [24]. Several studies validated the 18k SNP set for the evaluation of genetic diversity in V. vinifera [19,23,24,26,[31][32][33][34], but the Vitis18kSNP array could also represent a potentially effective tool for rootstock characterization, due to the consistent number of SNPs detected in the Northern American species genome.…”

“…vinifera ) cultivars account for 50% of the totally used acreage for commercial production (OIV 2017). Within these predominant cultivars, intra-varietal genetic variation, caused by somatic mutation (De Lorenzis et al 2017), is exploitable to select clonal variants of the desired cluster architecture phenotype.…”
