Abstract. The present investigation was aimed at developing and optimizing a simple aqueous tabletcoating formulation and its process. 5-Fluorouracil (5-FU) was used to ascertain the relative lipophilic/ hydrophilic behavior of the coating system. Optimization was performed by evaluating the adhesive force strength and cohesive force strength of the tablet coat using a texture analyzer. The in vitro release of 5-FU was found to decrease with an increase in (tablet surface-coat) adhesive force strength. The (tablet-tablet) cohesive force strength was reduced by the addition of magnesium silicate to the coating solution. The addition of magnesium silicate (0.2% w/v) to the carboxymethyl Cassia fistula gum-chitosan (CCG-CH) coating surface significantly inhibited the release of 5-FU possibly due to an increase in the hydrophobic character of the coated tablet surface. This was possible by coating cohesive force strength reduction coating compositions (CCG-CH (70:30) and 0.3% magnesium silicate). Further, the FTIR-ATR and DSC analyses suggested the pivotal role of magnesium silicate in modifying the release of 5-FU from CCG-CH-coated tablets due to hydrogen bonding of its Si-O-Si or Mg-O groups with -OH moieties of CCG-CH.KEY WORDS: 5-fluorouracil; carboxymethyl Cassia fistula gum; chitosan; magnesium silicate adhesive force strength of tablets; texture analysis.

Abstract. Precipitated calcium carbonate (PCC) was prepared by means of semicontinuous carbonation of Ca(OH) 2 suspension, at 35 and 45 °C and in the presence of non-ionic dextran and dextran sulfate. The carbonation process was regulated at different predetermined values of electrical conductivity, that corresponded to different concentrations of dissolved Ca(OH) 2 : 0.5 mS cm. The results of physicalchemical characterization of the product showed that calcite was the only polymorphic modification obtained in the whole range of experimental conditions investigated. In addition, it was found that morphology, crystal size distribution and specific surface area of PCC strongly depended on the electrical conductivity / concentration of dissolved Ca(OH) 2 , at which the process was performed: predominantly scalenohedral crystals of higher surface area (about 5 m 2 g −1) were produced at higher electrical conductivity, while at lower electrical conductivity predominantly rhomohedral calcite crystals of relatively low specific surface area were obtained. In the system of the highest electrical conductivity, κ 25 = 5.0 mS cm, and at 35 °C, the addition of non-ionic dextran significantly influenced the process by preventing the regulation. The crystals that appeared were in the form of irregular aggregates of high specific surface area, S ≈ 29 m 2 g −1. FT-IR and TG analyses indicated that the non-ionic dextran was adsorbed onto the calcite surface, most probably by relatively strong and specific interactions between oxygen from the hydroxyl groups of dextran molecules and calcium ions from the crystal surface. On the contrary, the anionic dextran (dextran sulfate) exerted minor effects in the course of semicontinuous carbonation process and in the properties of the final product, PCC. However, the analysis of the precipitate indicated that dextran sulfate was adsorbed at the surfaces, most probably by the weak and non-specific electrostatic interactions. (doi: 10.5562/cca1746)

The aim of the present study was to minimize drug release in the upper gastro intestinal tract and target the colon using the principles of compression coating. Compression-coated tablets of ibuprofen were prepared by a direct compression method using locust bean gum (LBG) at 300, 250, 200, and 175 mg. Tablets were evaluated for their physicochemical properties and in vitro drug release. In vitro drug release studies were performed with and without rat caecal contents. In rat caecal contents, tablets showed enhanced drug release due to degradation of the LBG coating by colonic enzymes. The in vitro release studies in pH 6.8 phosphate buffer containing 2% w/v rat caecal contents showed the cumulative percentage release of ibuprofen after 26 h as 39.91 ± 0.05%, 53.21 ± 0.37%, 69.17 ± 0.19%, and 94.46 % ± 0.92%. Coating thickness and the amount of chitosan control the release rate. Formulations were best fitted with Korsmeyer-Peppas kinetics, and the mechanism of drug release was non-Fickian super case II transport. FTIR studies reveal there is no drug-polysaccharide interaction. The F 1 formulation is a promising system for drug targeting to the colon.
