Studies on the solution conformation of human thioredoxin using heteronuclear nitrogen-15-proton nuclear magnetic resonance spectroscopy

Abstract: The solution conformation of uniformly labeled 15N human thioredoxin has been studied by two-dimensional heteronuclear 15N-1H nuclear magnetic resonance spectroscopy. Assignments of the 15N resonances of the protein are obtained in a sequential manner using heteronuclear multiple quantum coherence (HMQC), relayed HMQC-correlated (COSY), and relayed HMQC-nuclear Overhauser (NOESY) spectroscopy. Values of the 3JHN alpha splittings for 87 of the 105 residues of thioredoxin are extracted from a variant of the HMQC… Show more

“…The acquisition and processing details for these experiments were given by Feng et al (1995). The backbone 3 JNHa coupling constants extracted from a 2D 1 H-15 N HMQC-J spectrum Forman-Kay et al, 1990) were also reported earlier (Feng et al, 1995). A 3D simultaneously 13 C, 15 N-edited ROESY-HSQC spectrum was recorded using similar acquisition parameters as for the corresponding NOESY-HSQC experiment in order to confirm that certain cross-peaks were due to cross-relaxation and not to chemical or conformational exchange.…”

“…The 3JHN_Hc ~ coupling constants were measured through the use of an HMQC-J experiment (Kay et al 1989;Forman-Kay et al 1990;Kay and Bax 1990). The data were collected with 1024 complex points in T 1 and T 2 by using TPPI (Marion and Wfithrich 1983).…”
