Studies on the Regulation of (p)ppGpp Metabolism and Its Perturbation Through the Over-Expression of Nudix Hydrolases in Escherichia coli

Abstract: Stringent response mediated by modified guanosine nucleotides is conserved across bacteria and is regulated through the Rel/Spo functions. In Escherichia coli , RelA and SpoT proteins synthesize the modified nucleotides ppGpp and pppGpp, together referred to as (p)ppGpp. SpoT is also the primary (p)ppGpp hydrolase. In this study, using hypomorphic relA alleles, we provide experimental evidence for SpoT-mediated negative regulation of the amplification of RelA-depen… Show more

“…ApaH degrades dinucleoside polyphopshates, such as diadenosine tetraphosphate (Ap4A), a member of a second alarmone class in bacteria (Figure 4, A1) [44] . Interestingly, ApaH would be the first non‐Nudix type hydrolase interacting with MSN, while several Nudix‐type hydrolases have been described [21, 45–47] . We therefore investigated, if MSNs influence the activity of ApaH to uncover potential crosstalk between these different alarmones.…”

“…Deletion of the T. thermophilus ndx8 hydrolase results in ppGpp accumulation during exponential growth ( 74 ). Overexpression of the E. coli Nudix hydrolase genes nudG or mutT can complement deletion of the spoT hydrolase gene to relieve alarmone-mediated growth suppression ( 76 ). The E. coli Nudix proteins appear to deplete cytoplasmic pppGpp more aggressively than ppGpp.…”
