Studies on the Quantitative Autoradiography. I. Radioluminography for Quantitative Autoradiography of 14C.

Abstract: Evidence is presented to elucidate whether or not quantitative analysis of autoradiographs can be obtained with a 14C-labelled compound by the use of a new type of radiosensor called Imaging plate (IP). Our results showed that a linear relationship between relative intensity of PSL-BG and radioactivity of a given area was obtained. The linearity was maintained in a relatively wide range from 10(1) to 10(5) dpm of radioactivity. About 100 times higher sensitivity of the IP than any commercially available X-ray … Show more

“…The PCR product including the G2215A site was digested with HaeII at 37°C for 12 h. The digested DNA fragments were separated on an 8% PAG (Fig. 1E), and the dried gel was analyzed by a computerized system for radioluminography (BAS2000, Fuji Photo Film, Kanagawa, Japan; Motoji et al 1995), with analyzing software MacBAS version 2.3 (Fuji Photo Film; Fig. 1F).…”

“…IP used in this technique was developed by Fuji Photo Film Company in 1988 as a two-dimensional sensor capable of receiving and storing radiant energy [1]. The IP was confirmed to have linear characteristics within a range of 10 1 to 10 5 photostimulated luminescence -background (PSL-BG) units for both 3 H and 14 C [2,3]. Here, 100 PSL/mm 2 is defined to the output from the BAS system obtained when IP is irradiated by X-ray (80 kV, 0.15 mR) with W target.…”

“…Subsequently, the dried sections were exposed overnight to an imaging plate (BAS SR2040, 20 × 40 cm 2 ; Fujifilm, Tokyo, Japan). The imaging plate was then scanned with a phosphor image reader (FLA-5100; Fujifilm, Tokyo, Japan), and the obtained images were analysed using Multi Gauge 3.0 software (Fujifilm, Tokyo, Japan) to calculate the striatal specific uptake ratio (SUr), which is defined as (PSL striatum − PSL cerebellum )/PSL cerebellum [41], where PSL is the photostimulated-luminescence intensity [42, 43]. Five consecutive brain sections from each animal were used to determine the target PSL.…”
