Studies on the human T cell receptor α/β variable region genes. I. Identification of 7 additional Vα subfamilies and 14 Jα gene segments

Abstract: The anchored-polymerase chain reaction has been used to study further the diversity of the human T cell receptor alpha chain. The analysis of 308 cDNA transcripts from human peripheral lymphocytes hybridizing with a C alpha probe led to the identification of a series of additional V alpha and J alpha gene segments. The sequences of seven V alpha gene segments which individually define a novel V alpha subfamily (termed V alpha w23 to V alpha w29) are reported. The sequences of some previously described V alpha … Show more

“…AP511 has been described to contain an in-frame stop codon but, in our analysis, there is a point mutation that has changed this segment into a functional one. Ja33 to Ja36, Ja38, Ja4l, and Ja42 correspond to IGRJaO8, IGRJa12, IGRJaO3, IGRJaO5, IGRJaOlC, IGRUaM4, and IGRJaO4 (38); Ja37, Ja39, and Ja40 have not been described, to our knowledge. The V81 gene segment has been described (39).…”

“…Va21 sequences differed at residues 75 and 76, a change that reintroduces the highly conserved leucine at position 75 [MI. In Va29 two new variants were detected at positions 57-60 [21].The WR and JM Va29 differed by four nucleotides from the previously reported sequence; the AJ variant had only three changes. Finally, subject JM apparently has two variants of Va28, which differed at position 87 [21].…”

“…2 a. The a chain expression of these clones was also determined, by sequencing the products of a chain-specific anchor PCR. Once more the five clones fell into two categories: the Vp5.2-expressing clones MAW1.4, 1.5 and 1.8 each expressed a member of the Va2 family highly related toVa2.3 [42] which we have called Va2.4,whereas theVP5.6-expressing clones MAW10.8 and 10.15 expressed a member of the Vaw23 family, [43] which we have designated Vaw23.2. In the case of MAW10.8, shown to be biclonal with respect to P chain expression, a second a chain was also detected.…”
