SUMMARYA herpesvirus was isolated from a lymphosarcoma culture of tree shrews (Tupaia belangeri) and termed Tupaia herpesvirus 2 (THV-2). Electron microscopy of THV-2 revealed the presence of virus particles with nucleocapsids of about Ioo nm surrounded by large envelopes compatible with virions of the herpesvirus group. An extensive host range study revealed the Tupaia embryonic fibroblasts are the cells of choice for the efficient propagation of THV-2. This cell line was used for the continued propagation and plaque assay of THV-2. The tool. wt. of the virus D N A was found to be too x lot The buoyant density of THV-2 was 1-724 g/ml. The DNA of THV-2 was compared to the DNA of herpes simplex virus (HSV) and to another previously isolated herpesvirus from apparently healthy tree shrews (THV-I). The analysis was carried out using the restriction endonuclease EcoRI and the cleavage pattern of THV-2 D N A resulted in DNA fragments which were different from those ofHSV-I DNA and distinguishable from the DNA of THV-I.

(Received November 17th, 1976) Summary A physical map of Neurospora crassa mitochondrial DNA has been constructed using specific fragments obtained with restriction endonucleases. The DNA has 5 cleavage sites for endonuclease Bam HI, 12 for endonuclease Eco RI and more than 30 for endonuclease Hind III. The sequence of the Eco RI and Barn HI fragments has been established by analysis of partial fragments. By digestion of the Eco RI fragments with Barn HI, a complete overlapping map has been constructed. The position of the largest Hind III fragment on this map has also been determined. The map is circular and the added molecular weight of the fragments is 40 • 106, which is in good agreement with earlier measurements on intact DNA, using the electron microscope.
