Drug discovery often begins with the screening of large compound libraries to identify lead compounds. Recently, the enzymes involved in the biosynthesis of natural products have been investigated for their potential to generate new, diverse compound libraries. There have been several approaches toward this end, including altering the substrate specificities of the enzymes involved in natural product biosynthesis and engineering functional communication between enzymes from different biosynthetic pathways. While there exist assays to assess substrate specificity of enzymes involved in these pathways, there is no simple method for determining whether enzymes from different synthases will function cooperatively to generate the desired product(s). Herein we report a method which provides insight into both substrate specificity and compatibility of protein-protein interactions between the acyl carrier protein (ACP) and ketosynthase (KS) domains involved in fatty acid and polyketide biosynthesis. Our technique uses a one-pot chemoenzymatic method to generate post-translationally modified ACPs capable of covalently interacting with KS domains from different biosynthetic systems. The extent of interaction between ACPs and KSs from different systems is easily detected and quantified by a gel-based method. Our results are consistent with previous studies of substrate specificity and ACP-KS binding interactions and provide new insight into unnatural substrate and protein interactions.

We primarily optimize the ground‐state crystal structure, obtaining Cu‐N1 bond length, λ Cu‐N1 = 2.00 Å, N1‐N2 bond length, λ N1‐N2 = 1.23 Å, N2‐N3 bond length, λ N2‐N3 = 1.17 Å, ∠Cu‐N1‐N2 bond angle, ϕ = 124.4 ° , and ∠N1‐N2‐N3 bond angle, θ = 178.6 ° , which agree with the reported experimental values . The crystal Cu(N 3 ) 2 shows the ferromagnetic state and the calculated magnetic moment μ = 0.184 μ B.…”

Amorphous polymerization of nitrogen in compressed cupric azide

Metal azides have attracted increasing attention as precursors for synthesizing polymeric nitrogen. In this article, we report the amorphous polymerization of nitrogen by compressing cupric azide. The ab initio molecular dynamics simulations show that crystalline cupric azide transforms into a disordered network composed of singly bonded nitrogen at a hydrostatic pressure of 40 GPa and room temperature. The transformation manifests the formation of a π delocalization along the disordered Cu‐N network, thus resulting in a semiconductor–metal transition. The estimated heat of formation of the amorphous polymeric nitrogen system is comparable to conventional high‐energy‐density materials. The amorphization provides an alternative route to the polymerization of nitrogen under moderate conditions.

The reaction of the low melting solid PPN(N3) with copper(II) acetate in ethanol yields (PPN)2[Cu11(N3)24(C2H5OH)2]. The undecanuclear azidocuprate dianon is unprecedented and represents the largest example for a discrete azidocuprate(II) anion to date. This structurally more‐or‐less flat anion contains features of Cu(N3)2, i. e. can be considered the largest so far known building block of this solid.
