Studies in Experimental Animals on Disseminated Intravascular Coagulation (DIC)

Abstract: SummaryChanges in the clotting system, as well as morphological and functional alterations corresponding to that of the pathologic phenomenon of disseminated intravascular coagulation (DIC) or consumption coagulopathy, were produced by thrombin infusion (550 NIH U × kg−1 × h−1) in rats and simultaneous inhibition of fibrinolysis by PAMBA (100 mg/kg).Changes in the fibrinogen level and platelet count as well as the appearance of fibrin monomers and the formation of microthrombi in several organs were evaluated.… Show more

“…DIC was induced by thrombin infusion as desc, ribed elsewhere (16). Two hr before the infusion the rats were pretreated with tzsl-g6rinogen (0.5 MBq) i. v. In ethyl urethane anaesthesia a catheter was inserted into the dissected right jugular vein up to the right ventricle and thrombin (550 NIH-Uftg ' h) was infused.…”

“…There is a wide variety of experi mental models that facilitate studies on the pathogenesis of disseminated intravascular coagulation (DIC) [4], Normally, clotting en zymes and bacterial endotoxins were used as triggers, but in special investigations, bio genic amines were also employed. Rats, rab bits, dogs and pigs served as experimental animals [5][6][7][8], For pharmacological studies on the influence of recombinant hirudin (r-hirudin) on DIC we used animal models which allow to continuously follow altera tions in the clotting variables and the forma tion of microthrombi [9].…”

“…A fluorescent antibody technic may be needed to identify the fibrin [55]. When thrombin is injected into a rat, fibrin is first deposited in the lung and only later in the liver, kidney, and adrenal [145]. In the various animal studies, fibrin deposition was minimized by agents such as heparin, dipyridamole, and trysylol (Trasylol).…”
