Structure of the murine tissue factor gene. Chromosome location and conservation of regulatory elements in the promoter.

Abstract: Tissue factor (TF) is a transmembrane glycoprotein that mediates cellular initiation of the coagulation serine protease cascades. Moreover, expression of TF in human atherosclerotic plaques is likely to play a significant role in the thrombotic complications associated with plaque rupture. In this study the complete murine TF gene, Cf-3, was isolated from mouse NIH 3T3 cells and was found to consist of six exons spanning about 11 kilobase pairs (kbp) of DNA. A major transcriptional start site was located 24 bp… Show more

“…Two dierent AP1-dependent promoter constructs were tested. AP1-TF60 consists of the TF AP-1 elements linked to the mouse TF promoter deleted to 760 and containing only the TATA box as a recognizable sequence motif (Felts et al, 1995;Mackman et al, 1992). AP1-tk consists of the same TF AP-1 elements linked to the heterologous tk promoter.…”

“…One of the key regulatory molecules in this regard, tissue factor, has been shown to contain NF-B regulatory element(s) in its promoter region, along with other transcription factor binding sites, such as SP-1 and Egr-1. 42,43 Tissue factor is expressed on the EC surface in response to cytokines (IL-1, TNF␣) or LPS. Expression of dnIKK2 inhibited clot formation in LPS-stimulated HUVECs, demonstrating that inhibition of NF-B is sufficient to prevent hemostasis in this coagulation assay.…”

“…Mouse TF mRNA was detected using forward primer spanning exon 4 and 5 (5Ј-TCAAGCACGG-GAAAGAAAAC-3Ј) and reverse primer located within exon 5 (5Ј-CTGCTTCCTGGGCTATTTTG-3Ј), which generated a 137 base pairs product. 37 TF expression was normalized to the expression of hypoxanthineguanine phosphoribosyltransferase (forward, 5Ј-GTGGTGAAAAGGAC-CTCTCG-3Ј and reverse 5Ј-TGAAGTACTCATTATAGTCAAGGGGA-3Ј), and relative expression levels were calculated using the comparative Ct method.…”
