Structure of t cell antigen receptor β chain in synovial fluid cells from patients with rheumatoid arthritis

Abstract: We attempted to identify a clonal proliferation of T cells from synovial fluid samples from patients with rheumatoid arthritis, using techniques of restriction fragment length polymorphism. We used probes for the Rheumatoid arthritis (RA) is thought to develop as a consequence of disordered immune regulation in a genetically susceptible host (1). Although no inciting agent has been detected, evidence has accumulated implicating an infectious etiology-possibly viral. Moreover, data support the concept of a loca… Show more

“…Direct sequencing of selected V-J combinations confirmed that these Gaussian distributions did not contain an expanded clonal specificity which exceeded 25% of a particular V-J combination and could therefore be detected by direct sequencing. In contrast, deviations from the Gaussian distribution and dominance of one particular CDR3 length was seen for 12 V,/-Jf3 combinations in the three patients (Fig. 4).…”

“…It has been proposed that mycobacterial antigens are important in RA since mycobacterial reactive clones have been isolated from synovia (31). However, synovial T cells do not always appear to be clonally restricted in their T-cell receptor usage and this argues against a single local antigen response (32)(33)(34)(35)(36)(37). Lymphocytes can also reach inflammatory sites in other ways: by recognition of inflammatory endothelium (26) or by chemotaxis toward factors released by activated cells.…”

“…Evidence of oligoclonality manifested by the presence of rearranged bands detected by Southern blotting was rarely found in T cells directly isolated from synovial fluid samples (16,17). Moreover, T cell clones established in culture conditions designed to allow outgrowth ofall cells rarely exhibited identical rearrangements (18,19).…”
