Structure, mapping and expression of a growth factor inducible gene encoding a putative nuclear hormonal binding receptor.

Abstract: We have characterized a growth factor inducible gene, N10, encoding a nuclear protein of 601 amino acids with a significant similarity to members of the steroid and thyroid hormone receptor families. The gene is rapidly but transiently induced by several mitogens. Immunoprecipitation studies show that the N10 protein is transiently expressed after stimulation of quiescent cells, presenting a half‐life of approximately 30 min. The N10 transcription unit is 8 kb in length, split into seven exons. The exon‐intron… Show more

“…Nr4a1 (also known as nur77 and NGFI-B) belongs to a subfamily of orphan nuclear receptors, whose other members are Nr4a2 [also known as Rnr-1 (ribonucleotide reductase-1), Not, and Nurr1] and Nr4a3 [also known as Nor-1 (neuron-derived orphan receptor-1) and Tec], all of which function as transcription factors (Ryseck et al, 1989;Steinmetz et al, 2001). Several studies have implicated Nr4a1 and Nr4a2 as CREB target genes (Fass et al, 2003;Darragh et al, 2005;von Hertzen and Giese, 2005ab).…”

“…The apoptotic process is activated by the expression of Nur77, an orphan steroid receptor [67][68][69]. Constitutive expression of Nur77 in thymocytes results in a dramatic involution of the thymus, whereas expression of a dominant-negative Nur77 interferes with negative selection [70,71].…”
