Structure determination ofStreptococcus suisserotype 14 capsular polysaccharide

Abstract: The capsular polysaccharide (CPS) of Streptococcus suis serotype 14 was purified, chemically modified, and characterized. Sugar and absolute configuration analyses gave the following CPS composition: D-Gal, 3; D-Glc, 1; D-GlcNAc, 1; D-Neu5Ac, 1. The Sambucus nigra lectin, which recognizes the Neu5Ac(α2-6)Gal/GalNAc sequence, showed binding to the native CPS. Sialic acid was found to be terminal, and the CPS was quantitatively desialylated by mild acid hydrolysis. It was also submitted to periodate oxidation fo… Show more

“…From 8 liters of GBS type III or type V culture, 360 mg or 600 mg initial crude capsule was obtained, respectively, to afford around 30 mg of purified CPS after gel filtration (average yield of 3.75 mg CPS/liter of culture). GC and NMR analyses of purified CPSs gave sugar compositions and structures in accord with previous findings (8,10,11) (Table 1; see Fig. S1 and S2 in the supplemental material).…”

“…The cells were pelleted by centrifugation at 10,000 ϫ g for 40 min, suspended by repeated pipetting in 33 mM phosphate-buffered saline (PBS), pH 8.0, and chilled. The CPSs were then purified as previously described (10,11).…”

“…However, no cross-reaction was reported between the serotype 14 reference strain and anti-serotype 1 serum (7), indicating that some differences in the CP structures certainly exist between the two serotypes. Indeed, Elliott and Tai showed that the serotype 1 CP contains N-acetyl galactosamine (43), but the sugar was not present in the serotype 14 CP structure determined by Van Calsteren et al (46). It is noteworthy that, in serotype 1 reference strain NCTC 10237 and in another strain (strain ST1, accession number CP002651), the genes corresponding to a glycosyltransferase gene (cps14G) of the serotype 14 reference strain seem to be disrupted by a single base deletion and the resultant frameshift mutation (see Fig.…”

“…However, a few strains were weakly hydrophobic, implying that they have completely new cps gene clusters. So far, the CP structures have been determined only for serotypes 2 and 14 (20,21), and cps gene clusters of most of the nonserotypeable isolates have not been analyzed yet. Further analysis, including structure determination of the CPs and cps gene clusters of S. suis strains with different antigenicity, may provide insights into the evolution of S. suis CP as well as the relationship between cps types and serotypes.…”
