The rpN5, rpl20, rplS. rps8, and a portion of the rp16 genes of the cyanelle genome of Cyanophora paradoxa have been cloned, mapped and sequenced. Homologs of the rpl35, rpi5, and rp16 genes are not found in the chloroplasts of higher plants. The rpi35 and rpf2@ genes most likely form a dicistronic operon which is located upstream from the apeE-apcA-apeE locus of the cyanelle and which is divergently transcribed from this locus. The rpfJ, rpl8, and rpl6 genes probably form a part of a larger cluster of genes encoding components of the cyanellar ribosomes. These genes are organized in a fashion similar to that observed in all procaryotes examined to date, with the exception that the rpsI4 p;ene is not found between the rpi5 and rps8 coding sequences. Hypotheses concerning the origins of cyanelles and chloroplasts are discussed.

sec Y is a prokat~otic gone that encodes the SeeY protein, an integral membrane component of the prokaryotic protein translocation apparatus. A chloroplast-encoded see Y homologue has been identified in the unicellular, chromophytic alga, Pavlova lutherii. The gone predicts a protein composed of ten membrane-spanning regions, that is approximately 25% homologous and 50% similar to bacterial and plastid S¢¢Y proteins. The secl' gone from P. lutherli is independent of the ribosomal protein (rp) gone cluster to which it is closely linked in other organisms. In P. Iutherii see}" is located 5' to atpl and atpH. Since, in higher plants the atplHFA gone cluster and the rp gen0 cluster are s~paratcd by approximately 50 kb, we conclude, this indicates a novel chloroplast gone arrangement in P. lutherii.

We have isolated mutants of Synechocystis PCC6803 that grew very slowly in a low-sodium medium, which is unfavorable for HCO3 transport, and examined two of these mutants (SC1 and SC2) for their ability to take up CO2 and HCO in the light. The COz transport activity of SC1 and SC2 was much lower than that of the wild type (WT), (18) supplemented with 20 mM Hepes-KOH buffer (KOH added to pH 8.0) during aeration with 3% vol/vol CO2 in air. Low-sodium medium was prepared by adding NaCl (final concentration, 100 ,tM) to the BG11 medium in which all of the sodium salts were replaced by potassium salts. Continuous illumination was with tungsten lamps providing 120 tumol of photosynthetically active radiation/m2.s. Cells in a logarithmic phase of growth were mutagenized with N-methyl-N'-nitro-N-nitrosoguanidine (40 ag/ ml) for 20 min in the light, washed twice, and grown with 3% C02/97% air in BG11 medium. Cells were then grown for 2 days with air in the presence of ampicillin (100 ,tg/ml) in the low-Na+ (100 tLM) BG11 medium. The activity of HCO3 uptake in Synechocystis PCC6803 at this Na+ concentration was much lower than at 15 mM Na+ (see curves a and e in Fig. 2C
