Structure and expression of a human oxytocin receptor

Abstract: Just before the onset of labour, uterine myometrium becomes extremely sensitive to oxytocin, for which it is a primary target tissue, because of a dramatic increase in the number of oxytocin receptors. We report here the structure and expression of the human oxytocin receptor complementary DNA isolated by expression cloning. The encoded receptor is a 388-amino-acid polypeptide with 7 transmembrane domains typical of G protein-coupled receptors. The oxytocin receptor, expressed in Xenopus oocytes, specifically … Show more

“…Effects of OT in the CNS and in the periphery (e.g., heart and cardiovascular system, kidney, reproductive organs) are critically dependent on presence of OT receptors (OTR) (Gimpl and Fahrenholz, 2001). The OTR is a 389-amino acid polypeptide and belongs to the G-protein coupled receptor superfamily (Gimpl and Fahrenholz, 2001; Kimura et al, 1992). The OTR expressing gene ( OXTR ) is located on chromosomal region 3p25–3p26.2, spans 17 kb, and consists of 3 introns and 4 exons (Gimpl and Fahrenholz, 2001).…”

“…4A(À)). PGF 2a and oxytocin are involved in pregnancy and parturition (Casey & MacDonald 1997;Sugimoto et al 1997), and both receptors are expressed in the ovary (Kimura et al 1992;Sugimoto et al 1994;Ezashi et al 1997), PGF 2a and then oxytocin were added to the hCG-treated granulosa cells. As shown in Fig.…”
