Structurally related (−)-epicatechin metabolites in humans: Assessment using de novo chemically synthesized authentic standards

Abstract: Accumulating data suggest that diets rich in flavanols and procyanidins are beneficial for human health. In this context, there has been a great interest in elucidating the systemic levels and metabolic profiles at which these compounds occur in humans. Although recent progress has been made, there still exist considerable differences and various disagreements with regard to the mammalian metabolites of these compounds, which in turn are largely a consequence of the lack of availability of authentic standards … Show more

“…This is in line with the notion of altered metabolism, accumulation, and excretion of certain metabolites and uremic solutes in ESRD (48). Recent data suggest that the elimination of CF is dependent on the urinary excretion of their corresponding metabolites (49). We now show that metabolites are dialyzable, which opens new insights into pharmacokinetics.…”

“…Despite reported differences in analytical procedures between laboratories [47], the consensus is that (−)-epicatechin-3 -O-glucuronide, (−)-epicatechin-3 -O-sulfate, and a 3 -Omethyl-(−)-epicatechin-5/7-sulfate are the predominant epicatechin metabolites in humans after oral consumption [47,49]. The apparent half-life of most epicatechin conjugates is 2-4 h…”

“…It undergoes sulfation, methylation and glucuronidation with typical T max for conjugates of ~2 h [46,47]. After both oral ingestion and direct jejunal perfusion, the EC-3 -O-glucuronide conjugate is the most abundant metabolite found in plasma [48] (Fig.…”

“…EC and other flavan-3-ols are extensively metabolized in the small intestine and in the liver in humans and rodents following oral consumption (4,37), which has generated questions as to whether the parent compound or bioactive metabolites are responsible for its beneficial effects. We and others have hypothesized an alternative mechanism of protection involving activation of the nuclear transcription factor erythroid 2p45-related factor-2 (Nrf2).…”
