Structural Studies on Teichoic Acids in Cell Walls of Several Serotypes of Listeria monocytogenes1

Abstract: Structural studies were carried out on the teichoic acids in cell walls of Listeria monocytogenes serotypes 3a, 4b, 4f, 6, and 7. The structure of the dephosphorylated repeating units, obtained by treatment with 46% hydrogen fluoride or alkaline hydrolysis, was examined by methylation analysis, acetolysis, and 1H-NMR spectroscopy. The results of Smith degradation of the teichoic acids and 13C-NMR spectroscopy led to the following most likely structures of the repeating units of the teichoic acids:----1-[N-acet… Show more

“…A total of 18 of 29 Listeria strains tested were lysed by P70, including food isolates, reference strains, and the novel species L. marthii (9) but not L. rocourtiae (17). Consistent with other Listeria phages isolated so far, P70 does not bind to or lyse serovar 3 strains that lack the rhamnose residue in the polyribitol phosphate backbone of wall teichoic acids (7,26).…”

“…The main components of the listerial cell wall are PG, LPS, and TAs (253). The WTAs of serovars 1/2, 3, and 7 are decorated with GlcNAc or Rha residues, while GlcNAc is incorporated into the backbone of the WTAs of serovars 4, 5, and 6, and the GlcNAc is again decorated with Glc and Gal residues (254).…”

“…Most of the descriptions of the composition and suspected structure of the L. monocytogenes membrane, peptidoglycan, and associated polymers were made about 20 to 40 years ago (57,65,171,180,181). However, this topic now is receiving further attention, which should help to refine the overall structure (for a review, see reference 145).…”
