Structural studies on lens proteins

Abstract: The sequence around the thiol group in lens proteins has been investigated. The proteins were converted into their carboxy[(14)C]methyl derivatives and submitted to partial acid hydrolysis, or digested with proteolytic enzymes. Acid hydrolysis of bovine alpha-crystallin gives N-seryl-(S-carboxymethyl)cysteine, Ser-CMCys (Waley, 1965a), but this dipeptide is not obtained from beta-crystallin or gamma-crystallin. Trypsin and chymotrypsin also give different peptides from the three crystallins. The radioactive pe… Show more

“…Although they have been studied for many years no structural comparison of them has been made. The amino acid sequence around the single cysteine residue of ce-crystallin is -Ser-Cys-Ser-Leu- (Mok & Waley, 1967). fl-Crystallin and y-crystallin were found to differ considerably from a-crystallin in the number and distribution of cysteine residues.…”

“…The sequence of the first 11 residues in the A chain had been determined (Corran & Waley, 1969), and this sequenee includes one methionine and one tryptophan residue. Limited sequences around the cysteine residue have also been reported (Waley, 1965;Mok & Waley, 1967); we now report an extended sequence of 28 residues around this residue. This sequence includes the second residue of methionine in the chain.…”

“…1), and this was confirmed by cleavage with cyanogen bromide. Of the other chymotryptic peptides in Table 2, peptide Cl was impure, but was derived from the N-terminal region, and peptide C4 from the C-terminal region; peptide C2 was the tetrapeptide isolated by Mok & Waley (1967). Another point, namely the order at the junction of residues 11-12, was confirmed by cleavage in acid.…”

“…ox-Crystallin. This was prepared from ox lenses as previously described (Mok & Waley, 1967). When the protein was to be for the preparation of A and B chaiiis, calf lenses were used, and the precipitation by (NH4)2S04 was omitted, and instead the protein was purified by chromatography on Sephadex G-200 (Schoenmakers et al 1968).…”
