Structural Studies on Heparan Sulphates

Abstract: Three heparan sulphate fractions were subjected to degradation by periodate oxidation and alkaline elimination. The starting materials were one low-sulphated fraction rich in glucuronic acid and iV-acetylglucosamine (I), one fraction (IV) that was medium-suphated, contained similar proportions of iV-acetyl and iV-sulphate as well as of glucuronic and iduronic acid and one fraction (V) that was oversulphated, contained an excess of iV-sulphate and was rich in iduronic acid. Selective periodate oxidation of gluc… Show more

“…We report the selective removal of GlcA- p NP moiety from a more structurally complex heparin-like oligosaccharide using both Smith degradation and alkaline elimination 2224 . This approach has been previously used for micro scale analysis of polysaccharides relying on mass spectrometry (MS) and leaves unnatural moieties at the reducing end of the resulting degradation products 2528 . We have modified this method to prepare a natural heparin pentasaccharide rich in trisulfated disaccharide units.…”
