Structural Refinement of the Tubulin Ligand (+)-Discodermolide to Attenuate Chemotherapy-Mediated Senescence

Abstract: The natural product (+)-discodermolide (DDM) is a microtubule stabilizing agent and potent inducer of senescence. We refined the structure of DDM and evaluated the activity of novel congeners in triple negative breast and ovarian cancers, malignancies that typically succumb to taxane resistance. Previous structure-activity analyses identified the lactone and diene as moieties conferring anticancer activity, thus identifying priorities for the structural refinement studies described herein. Congeners possessing… Show more

“…Studies using discodermolide, an anticancer drug with great similarities to dictyostatin, did not show improvement, despite initially promising results. Moreover, discodermolide showed high toxicity that led to pulmonary interstitial disease (Mita et al, 2004;Falkenberg et al, 2019;Guo et al, 2020).…”

“…Discodermolide, isolated from sponges ( Discodermia dissolute ), is the most promising natural product discovered to date. Discodermolide has been shown to be more effective than taxol and is being tested for the treatment of solid tumors ( 107 , 108 ).…”
