Abstract:In vitro growth assays with a purified o-galactose-specific lectin Kb-CWL I extracted from Kluyveromyces bulgaricus showed marked antifungal effects on 9 of 12 test strains belonging to the genera Kluyveromyces, Saccharomyces, Pichia, Candida, Rhodotorula and Schizosaccharomyces. The inhibition of growth was proportional to the lectin concentration (0.04 and 0.08 mg ml-l) in the culture medium. Under the test conditions, K bulgaricus, I£ lactis and S. bayanus were consistently agglutinated without growth inhibition. D-Galactose, the inhibitory sugar of the cell aggregation activity, did not abolish the antifungal effects of the lectin Kb-CWL I on other yeast strains tested. In C. albicans and C. tropicalis, the lectin influenced the dimorphism of these strains and stimulated germ tube formation.

Rksumk : Deux souches, l'une floculante, l'autre non floculante, de la levure Kluyveromyces luctis ont kt6 cultivies dms un milieu liquide de Sabouraud avec Ca 0,Q7 mM. Le traitement B l'ithylknediamine des parois isolkes sCpare trois fractions : A, B et C. La fraction A, soluble dans l'Cthylknediarnine, est fomCe de polymkres hydrosolubles de nature phosphopeptidomannane. Ces derniers constituent la couche externe de l'architecture pariCtale. Les phosphopeptidomannmes ont Ct C Cgalement extraits en traitant les levures entikres B l'autoclave B 140°C dans un tampon citrate B pH 7. L'ktat de floculation et celui de non-floculation de ces levures se traduit par des variations quantitatives et structurales de ces phosphopeptidomannanes. Chez les levures floculantes, les parois contiennent une teneur plus ClevCe de ces polymkres pCriphCriques dont la masse molCculaire est plus importante que chez les levures non floculantes. Ceci rCsulte d'une structure plus complexe, en particulier de la prksence d'un plus grand nombre de ramifications B trois et quatre unitCs de mannose. L'analyse des produits d'acCtolyse rCvkle, en effet, que les phosphopeptidomannanes des levures floculantes libkrent essentielleinent des unites de mannotriose et de mannotCtraose phosphorylCes, alors que les polymkres des levures non floculantes se caractkrisent par la prksence d'unitCs de mannobiose. Lorsque ces polymkres sont soumis B une rkaction de p-elimination, ils libkrent des unitCs de mannobiose et de mannose dans un rapport qui fait apparaitre davantage d'unitks de mannobiose chez la levure floculante. En utilisant une lectine extraite de la souche floculante, et en l'incubant avec des Crythrocytes activCs par des phosphopeptidomannanes issus de levures floculantes et non floculantes, il apparait nettement que les Crythrocytes les plus agglutinCs sont ceux activCs par les polymkres issus des levures floculantes. La fraction C, insoluble dans l'ithyl2nediamine, correspond B la matrice rigide de la paroi. L'Ctude de sa composition chimique ne fait pas apparaitre de diffkrences significatives entre la souche floculante et la souche non floculante.Abstract: Two strains, flocculating and nonflocculating, of the yeast Kluyveromyces luc'tis were grown in a Sabouraud's liquid medium containing 0.07 mM Ca. Treatment by ethylenediamine of isolated cell walls yielded three fractions: A, B, and C. Fraction A, soluble in ethylenediarnine, contained phosphopeptidomannan-like hydrosoluble polymers; these constituted the external wall of the parietal layer. Phosphopeptidomannans have also been extracted from entire yeast cells autoclaved at 140°C in a citrate buffer at pH 7.0. The flocculating and nonflocculating states of yeasts showed structural and quantitative variations in phosphopeptidomannans. The walls of the flocculating yeast cells contained higher amounts of peripheral polymers, the molecular masses of which were greater than those of nonflocculating yeast cells.
