Structural insights into repression of the Pneumococcal fatty acid synthesis pathway by repressor FabT and co‐repressor acyl‐ACP

Abstract: The Streptococcus pneumoniae fatty acid synthesis (FAS) pathway is globally controlled at the transcriptional level by the repressor FabT and its co‐repressor acyl carrier protein (acyl‐ACP), the intermediate of phospholipid synthesis. Here, we report the crystal structure of FabT complexed with a 23‐bp dsDNA, which indicates that FabT is a weak repressor with low DNA‐binding affinity in the absence of acyl‐ACP. Modification of ACP with a long‐chain fatty acid is necessary for the formation of a stable complex… Show more

“…Meanwhile, oligonucleotide 21 plus oligonucleotide 22 was a palindromic sequence with high AT content, suggesting that RcrR prefers binding to DNA regions with high AT content. This preference was also similar to those of the MarR family regulators CouR ( 26 ) and FabT ( 27 ).…”

“…SZU445, FAS (KEGG, EC: 2.3.1.86) (total gene length: 12,443 bp; CDS length: 12,285 bp; number of amino acids: 4147; number of copies: 1) and acyl carrier protein (ACP) (COG, COG ID:COG0236) (total gene length: 429 bp; CDS length: 429 bp; number of amino acids: 143; number of copies: 1) were sequenced and annotated. In the FAS pathway, the acetic acid to long-chain saturated fatty acids (C16:0 and C18:0) step of primary fatty acid biosynthesis is catalyzed by a type I FAS, and long-chain unsaturated fatty acids are generated after a string of desaturation and elongation reactions through desaturases and elongases [27]. FAS is a multifunctional polypeptide of 4147 amino acids that includes four catalytic domains for biochemical reactions: condensation, ketoacyl reduction, hydroxyacyl dehydration, and enoyl reduction [28].…”

“…The wings of bsCggR DBD (residues 64-73) support the protein-DNA interaction by wedging into the adjacent minor grooves. This function of the wings is very often observed in wHTH proteins (Zuo et al, 2019;Fillenberg et al, 2015;Morgan et al, 2019).…”
