Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) is an excellent tool for bacterial identification. It allows high throughput, sensitive and specific applications in clinical diagnostics and environmental research. Currently, there is no optimal standardized protocol for sample preparation and culture conditions to profile bacteria. The performance of MALDI-TOF MS is affected by several variables, such as sample preparation, culture media and culture conditions, incubation time/growth stage, incubation temperature, high salt content, blood in the culture media, and others. This review thus aims to clarify why a uniformed protocol is not plausible, to assess the effects these factors have on MALDI-TOF MS identification score, and discuss possible optimizations for its methodology, in relation to specific bacterial representatives and strain requirements.

The current study aimed to investigate exopolysaccharides (EPSs) produced by two Antarctic yeasts isolated from soil and penguin feathers samples collected on Livingston Island (Antarctica). The strains were identi ed as belonging to the species Leucosporidium yakuticum (LY) and Cystobasidium ongulense (CO) based on a molecular genetic analysis. The EPS production was investigated at submerged cultivation. Different chemical, chromatographic, and spectral analyses were employed to characterize EPSs. LY accumulated 5.5g/L biomass and 4.0g/L EPS after 120 h of cultivation, while CO synthesis 2.1g/L EPS at the end of cultivation and biomass amount reached 5.5g/L. LY-EPS was characterized by a higher total carbohydrate content (80%) and a lower protein content (18%) by comparison with CO-EPS (62%, 30%). The LY-EPS mainly consisted of mannose (90 mol%), whereas CO-EPS had also glucose, galactose, and small amounts of uronic acids (8-5 mol%). Spectral analyses (FT-IR and 1D, 2D NMR) revealed that LY-EPS comprised a typical β-(1→4)-mannan. Branched (hetero)mannan, together with β/α-glucans constituted the majority of CO-EPS. Unlike LY-EPS, which had a high percentage of high molecular weight populations, CO-EPS displayed a large quantity of lower molecular weight fractions and a higher degree of heterogeneity. LY-EPS (at a concentration of 100 ng/mL) elevated signi cantly interferon IFN-γ production in splenic murine macrophages and NK cells. The results indicated that newly identi ed EPS might affect IFN-γ signaling and in turn, might enhance anti-infectious responses. The data obtained also revealed the potential of EPSs and yeasts for practical application in biochemical engineering and biotechnology.

Discovery of a new bacterium, Microbacterium betulae sp. nov., in birch wood associated with hypersensitivity pneumonitis in woodworkers

A Gram‐positive, aerobic, rod‐shaped mesophilic bacterium was isolated from birch wood, referred to as the AB strain. Allergological tests suggest that this strain may cause allergic alveolitis in sawmill workers. Employing a polyphasic taxonomic approach, the AB strain's 16S rRNA gene sequence showed high similarity to Microbacterium barkeri and M. oryzae, with 97.25% and 96.91%, respectively, a finding supported by rpoB and gyrB sequence analysis. Further genome sequence comparison with the closely related M. barkeri type strain indicated a digital DNA–DNA hybridization value of 25.5% and an average nucleotide identity of 82.52%. The AB strain's cell wall peptidoglycan contains ornithine, and its polar lipids comprise diphosphatidylglycerol, phosphatidylglycerol, and unidentified glycolipids. Its major fatty acids include anteiso C15:0, anteiso C17:0, and iso C16:0, while MK‐10 is its predominant respiratory quinone. Comprehensive analysis through 16S rRNA, whole‐genome sequencing, phenotyping, chemotaxonomy, and MALDI‐TOF MS profiling indicates that the AB strain represents a new species within the Microbacterium genus. It has been proposed to name this species Microbacterium betulae sp. nov., with ABT (PCM 3040T = CEST 30706T) designated as the type strain.
