In this study, gradient ethanol precipitation method was applied to obtain the extracellular polysaccharides of Lactobacillus paracasei VL8 (VL8‐EPS). The yields, physicochemical properties, and immunomodulatory effects of VL8‐EPS obtained by precipitation at different ethanol concentrations (30%, 50%, and 70%, v/v) were compared. The results showed that VL8‐EPSs were high molecular weight sulfated heteropolysaccharides, composed mainly of glucose and galactose, and the alteration of ethanol concentration had an effect on their chemical compositions, molecular weight distributions, monosaccharide composition, and surface structure, while the primary structure remained the same. Among the three polysaccharide fractions, VL8‐EPS50 displayed better immunomodulatory activities compared with VL8‐EPS30 and VL8‐EPS70. VL8‐EPS50 was found to exert immunomodulatory effects by enhancing the phagocytic activity of RAW264.7 cells and to promote their secretion of more nitric oxide; it also showed stronger thermal and solution stability. In summary, there was a correlation between the structural characteristics of polysaccharides and their immunomodulatory activity, and VL8‐EPS50 was preferentially used for in vivo immunomodulatory activity. Practical ApplicationThis study opens up the source of raw materials for functional foods, which can provide some theoretical basis for the research and development of extracellular polysaccharides of lactic acid bacteria and promote their application in the future development of food industry.

The potential impacts of methanol extract from Polygonatum odoratum on (YZM) colonic histopathology, gut gas production, short-chain fatty acids (SCFAs), and intestinal microbiota composition were evaluated with dextran sulfate sodium (DSS)-induced colitis mice in this study. These results indicated that YZM increased colon length and ameliorated colonic histopathology in DSS-induced colitis mice. Moreover, YZM administration reversed intestinal microbiota compositions leading to the inhibition of H2S-related bacteria (e.g., Desulfovibrionaceae) and the lower level of H2S and higher contents of SCFA-related bacteria (e.g., Muribaculaceae). Taken together, the effects of methanol extract from Polygonatum odoratum are studied to provide new enlightenment and clues for its application as a functional food and clinical drug. Our study first revealed the relationship between intestinal gas production and key bacteria in ulcerative colitis.

Structural Characterization and Anti-breast Cancer Activity in vitro of a Novel Polysaccharide From Cymbopogon citratus

Cymbopogon citratus is an important functional food, widely used for flavoring in Africa and South America. In this study, a novel high-molecular-weight polysaccharide (CCP) from C. citratus was extracted, and its structural characteristics and anti-breast cancer activity in vitro were investigated. CCP contained both α and β configurations and mainly composed of galactose (36.89%), arabinose (23.97%), glucose (18.35%) and rhamnose (9.36%) with an average molecular weight of 1.98 × 106 Da. The main glycosyl residues of CCP detected by methylation analysis were 1,3,6-linked Galp, 1,3-linked Glcp, 1,5-linked Araf , T-Araf , and T-Rhap. In vitro experiments suggested that CCP significantly inhibited the proliferation of MDA-MB-231 cells, decreased the expressions of cyclin D1 and CDK4 and stocked cells at G0/G1 phase. Meanwhile, the typical morphological features of apoptotic cells were also observed. Combining with the consequences of Annexin V-FITC/PI staining, Hoechst 33258 staining and western blot analysis, CCP induced apoptosis of MDA-MB-231 cells by triggering the Fas/FasL-mediated death receptor pathway. Overall, these results provide a theoretical basis for the application of C. citratus polysaccharide as a potential anti-breast cancer agent in functional food and medicine.
