Plasmodium falciparum, the malaria-causing parasite, is a leading cause of infection-induced deaths worldwide. The preferred treatment approach is artemisinin-combination therapy, which couples fast-acting artemisinin derivatives with longer-acting drugs like lumefantrine, mefloquine, and amodiaquine. However, the urgency for new treatments has risen due to the parasite's growing resistance to existing therapies. Our study shows that a common characteristic of theP. falciparumproteome - stretches of poly-lysine residues such as those found in proteins related to adhesion and pathogenicity - can serve as an effective peptide treatment for infected erythrocytes. A single dose of these poly-basic peptides can successfully diminish parasitemia in human erythrocytes in vitro with minimal toxicity. The effectiveness of the treatment correlates with the length of the poly-lysine peptide, with 30 lysine peptides supporting the eradication of erythrocytic parasites within 72 hours. PEG-ylation of the poly-lysine peptides or utilizing poly-lysine dendrimers and polymers further increases parasite clearance efficiency and bolsters the stability of these potential new therapeutics. Lastly, our affinity pull-downs and mass-spectrometry identify P. falciparum's outer membrane proteins as likely targets for polybasic peptide medications. Since poly-lysine dendrimers are already FDA-approved for drug delivery, their adaptation as antimalarial drugs presents a promising new therapeutic strategy.

Present work stresses a novel analytical approach for increasing the specificity of standard NMR approaches for identifying polydimethylsiloxane (PDMS) and further silicone moieties in wines’ organic extracts, by including a second dimension that correlates chemical shifts with diffusion coefficients by means of pulsed-field gradient diffusion ordered spectroscopy (DOSY-NMR). Each silicone source in wines is unambiguously assigned by correlation of both local chemical environments and by a unique diffusion coefficient value, in turn related to a hydrodynamic radius (RH) that can be obtained with respect proper internal standards. Obtained PDMS diffusion coefficient values and hydrodynamic radii in wines’ extracts, in agreement with expected values, present a selectivity and specificity so far not reported, that positions DOSY-NMR spectroscopy as an alternative in oenology for controlling PDMS limits.

A composite comprising a silver nanoparticles‐loaded poly(amidoamine) (PAMAM) dendrimer crosslinked with glutaraldehyde (PAMAM/AgNPs) is synthesized using a one‐step method under vortex at room temperature. The structure of glutaraldehyde‐crosslinked PAMAM dendrimer is characterized using FTIR and high‐resolution magic angle spinning–carbon 13 nuclear magnetic resonance (HRMAS 13C NMR) spectroscopy. The crosslinking structure ensures the uniformity and stability of the Ag nanoparticles, and the flexibility of the PAMAM/AgNPs composite. X‐ray diffraction (XRD) analysis and energy dispersive spectroscopy (EDS) confirm the formation of Ag nanoparticles in the PAMAM/AgNPs composite. Scanning electron microscopy (SEM) and three‐dimensional (3D) micro‐Raman images reveal the coarsely porous 3D micro‐structure of the PAMAM/AgNPs composite. This unique micro‐3D architecture provides the PAMAM/AgNPs composite with spongy flexibility and a swab‐like nature, which is advantageous for the rapid and effective absorption of liquid samples on the surfaces of objects. Surface enhanced Raman spectroscopy (SERS) indicates that the detection limits of both rhodamine 6G (R6G) and thiram are 1.0 × 10−5 mol L−1, and the SERS enhancement factor of R6G is determined to be 1.35 × 104. The spongy flexible PAMAM/AgNPs SERS substrate therefore has potential for use in Raman active compound detection.
