The alpha replicons of the multi-replicon plasmids pGSH500 and pLV1402 have been characterized by DNA sequence analysis. Analysis of the DNA sequence of a 3672 bp HindIII fragment from pFDT100, which contains the pGSH500 alpha replicon, revealed similarity to a number of replicons belonging to, or related to, those of the IncFII family. The replicon region contains copA, tapA, repA and oriR, and replication initiation and termination sites are related to those from the IncFII replicon of R1. A copB gene was found to lie upstream of the HindIII site in the parental plasmid pGSH500. Downstream of oriR, a 707 bp region shows 726 % identity to a region of the Escherichia coli chromosome at 433', suggesting this region of pGSH500 may have been incorporated into the plasmid during a past chromosomal recombination event. Oligonucleotide primers homologous to consensus regions in the copB and repA genes, and the oriR regions from a number of IncFII-related replicons were used to amplify replication regions from pLV1402. Analysis of the amplified regions has shown the presence of copB, copA, tapA and repA genes. Phylogenetic analysis of Rep protein sequences from the RepFIIA family of antisense-control-regulated replicons revealed the presence of three distinct subgroups of Rep proteins. Comparative analysis of DNA and protein sequences from members of the RepFIIA family provides evidence supporting the roles of both non-selective divergence in co-integrate (multi-replicon) plasmids and Chi-mediatedrecombination in replicon evolution, and in particular, that such processes may have been widespread in the evolution of the RepFIIA family.

After incubation at 37°C in the absence of Ca21 ions, pathogenic strains of Yersinia spp. release large amounts of a set of plasmid-encoded proteins called Yops. The secretion of these proteins, involved in pathogenicity, occurs via a mechanism that involves neither the removal of a signal sequence nor the recognition of a C-terminal domain. Analysis of deletion mutants allowed the secretion recognition domain to be localized within the 48 N-terminal amino acids of protein YopH, within the 98 N-terminal residues of protein YopE, and within the 76 N-terminal residues of YopQ. Comparison of these regions failed to reveal any sequence similarity, suggesting that the secretion signal of Yop proteins is conformational rather than sequential. Hybrid proteins containing the amino-terminal part of YopH fused to either the a-peptide of 13-galactosidase or to alkaline phosphatase deprived of its signal sequence were efficiently secreted to the Yersinia culture medium. This observation opens new prospects in using Yersinia spp. as chimeric-protein producers and as potential live carriers for foreign antigens.

The Inc A/C plasmids, like Inc P and Inc Q plasmids, have a broad host range. However, their maintenance functions remain to be studied. An autoreplicative region of 2.79 kb named RepA/C, able to replicate both in the family Enterobacteriaceae and in Pseudomonas spp., was isolated and sequenced. The stability, copy number, and incompatibility expression of this replicon were determined. RepA/C and a nonautoreplicative fragment of 1.6 kb of this replicon were used as probes and showed specific hybridizations with the Inc P3-A/C plasmids from Pseudomonas spp. and members of the Enterobacteriaceae. These probes could be used as tools for identification of the plasmids of this epidemiologically important Inc group.Replication systems encoded by broad-host-range plasmids are generally more complex than those of narrow-host-range plasmids. In some cases the genes are scattered across several regions of the plasmid genome (35; see reference 15 for a review). Because of their large host range, these plasmids are of considerable interest both as natural vectors for disseminating genetic information among diverse species and as cloning vehicles (20).The maintenance of plasmids within bacterial populations requires both accurate replication and consistent distribution of plasmids to viable progeny (see reference 21 for a review). The replication and maintenance functions of the broad-hostrange plasmids Inc P (7, 24, 34) and Inc Q (9, 20) have been studied in detail. However, the maintenance functions of the Inc A/C plasmids have not been investigated. These plasmids are stably maintained in a variety of different hosts and are equivalent to the Inc P3 group in Pseudomonas spp. (12).We present herein the results of the isolation and the first molecular analysis of the RepA/C replicon specific to the Inc A/C group of members of the family Enterobacteriaceae and the Inc P3 group of Pseudomonas spp.

Abstract: We sequenced the minimum replication region of the virulence plasmid pYVe439-80 from a serogroup 0:9 Yersiniq enterocolitica. This sequence is 68% homologous on a 1,873-nucleotide stretch to the sequence of the RepFIHA replicon of the resistance plasmid R100.
