A preocupação com impactos ambientais tem estimulado muitos esforços visando substituir os combustíveis fósseis por biocombustíveis, como o etanol. Porém, para tornar isso possível, é necessário investir em outras tecnologias de produção, como o etanol de segunda geração (2G), de modo a elevar os níveis desse produto e assim atender à crescente demanda. Atualmente, esse tipo de produção ainda não é viável economicamente, devido aos altos custos dos coquetéis enzimáticos utilizados na etapa de sacarificação das biomassas lignocelulósicas. De modo a otimizar esses coquetéis, a busca por enzimas com atividades superiores tem sido visada. As βglicosidases, atuam na etapa final da sacarificação enzimática, catalisando a clivagem de ligações glicosídicas presentes na celobiose, liberando glicose e aumentando a eficiência das enzimas hidrolíticas que são inibidas pela celobiose no processo de hidrólise. Dessa forma, o projeto em questão realizou a clonagem, expressão em Pichia pastoris X-33, purificação e caracterização da enzima AfBgl1.3 de A. fumigatus. A avaliação estrutural da enzima recombinante, revelou a desestruturação da enzima devido ao aumento da temperatura. Na análise por dicroísmo circular (DC), foi possível observar a perda progressiva da estrutura αhélice e aumento da estrutura folha-β antiparalela, e permitiu determinar o valor de Tm aparente de 48,5 °C. Em adição, foi possível predizer a proporção das estruturas secundárias α-hélice (36,3%) e folha-β (12,4%), a 25 °C. A análise por emissão intrínseca do triptofano permitiu determinar a exposição dos resíduos apolares no ambiente externo da proteína, obtendo-se o valor de Tm aparente de 49,3 °C. A caracterização bioquímica sugeriu as condições ótimas para sua atividade sendo de pH 6,0 e temperatura de 40 °C. Nas temperaturas de 30 °C e 40 °C, a enzima apresenta-se estável, retendo cerca de 90 e 50% de atividade, respectivamente, após 24 horas de pré-incubação. A enzima também demonstrou alta estabilidade na faixa de pH 5-8, retendo mais que 65 % de sua atividade após 48 horas de pré-incubação. Como a inibição pelo produto é um dos desafios na utilização industrial de β-glicosidases, principalmente durante a degradação de celulose, avaliou-se a sua tolerância a glicose. A AfBgl1.3 recombinante foi estimulada em uma ampla faixa de concentração de glicose (1,8 -90 g L -1 ), aumentando cerca de 1,4 vezes sua atividade, na presença de 45 g L -1 de glicose. A enzima ainda apresentou atividade sobre os substratos salicina (495,0 ± 49,0 U mg -1 ), pNPG (340,5 ± 18,6 U mg -1 ), celobiose (89,3 ± 5,1 U mg -1 ) e lactose (45,1 ± 0,5 U mg -1 ), podendo ser classificada como de ampla especificidade. Os valores de Vmax encontrados para a AfBgl1.3, foram de 656,0 ± 17,5 U mg -1 , 706,5 ± 23,8 U mg -1 e 132,6 ± 7,1 U mg -1 , para os substratos pNPG, Salicina e celobiose, respectivamente. A enzima atuou em sinergia com o coquetel enzimático comercial (Celluclast ® 1.5L), uma vez que sua adição como suplemento, resultou em aumentos de cerca de 26% na taxa de conversão do...
