Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst) is one the most important diseases of wheat in Ethiopia and worldwide. To identify resistance genes, 90 bread wheat lines and 10 cultivars were tested at the seedling stage against one Pst race from Ethiopia and six races from China as well as evaluated for the stripe rust response in an inoculated field nursery at Yangling, Shaanxi province and in a naturally infected field in Jiangyou, Sichuan, China. Resistance genes were postulated using molecular assays for Yr9, Yr17, Yr18, Yr26, Yr29, Yr36, Yr44 and Yr62. Of the 100 entries tested, 16 had all stage resistance to all races. Molecular markers were positive for Yr9 in five genotypes, Yr17 in 21 genotypes, Yr18 in 27 genotypes, Yr26 in ten genotypes, Yr29 in 22 genotypes, Yr36 in 12 genotypes, Yr44 in 30 genotypes, and Yr62 in 51 genotypes. No line had Yr5, Yr8, Yr10 or Yr15. Complete or all stage resistance was observed in genotypes carrying gene combinations Yr9 ? Yr18 ? Yr44 ? Yr62, Yr29 ? Yr62 ? Yr26 and Yr9 ? Yr17 ? Yr26 ? Yr44 ? Yr62. The results are helpful for developing wheat cultivars with effective and more durable resistance to stripe rust both in China and Ethiopia.

Stripe rust or yellow rust (YR), caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most important diseases of wheat (Triticum aestivum L.). Widespread deployment of resistant cultivars is the best means of achieving durable disease control. The red grain, spring wheat cultivar Napo 63 produced by CIMMYT in the 1960s shows a high level of adult-plant resistance to stripe rust in the field. To elucidate the genetic basis of resistance in this cultivar we evaluated 224 F2:3 lines and 175 F2:6 recombinant inbred lines (RILs) derived from a cross between Napo 63 and the Pst-susceptible line Avocet S. The maximum disease severity (MDS) data of F2:3 lines and the relative area under the disease progress curve (rAUDPC) data of RILs were collected during the 2014–2015 and 2015–2016 wheat growing seasons, respectively. Combined bulked segregant analysis and 90K single nucleotide polymorphism (SNP) arrays placed 275 of 511 polymorphic SNPs on chromosome 2B. Sixty four KASP markers selected from the 275 SNPs and 76 SSR markers on 2B were used to identify a chromosome region associated with rust response. A major effect QTL, named Qyrnap.nwafu-2BS, was identified by inclusive composite interval mapping and was preliminarily mapped to a 5.46 cM interval flanked by KASP markers 90K-AN34 and 90K-AN36 in chromosome 2BS. Fourteen KASP markers more closely linked to the locus were developed following a 660K SNP array analysis. The QTL region was finally narrowed to a 0.9 cM interval flanked by KASP markers 660K-AN21 and 660K-AN57 in bin region 2BS-1-0.53. The resistance of Napo 63 was stable across all environments, and as a QTL, explained an average 66.1% of the phenotypic variance in MDS of F2:3 lines and 55.7% of the phenotypic variance in rAUDPC of F5:6 RILs. The short genetic interval and flanking KASP markers developed in the study will facilitate marker-assisted selection, gene pyramiding, and eventual positional cloning of Qyrnap.nwafu-2BS.

Background:Stripe rust, caused by the fungal pathogen Puccinia striiformis f. sp. tritici (Pst), is a serious foliar disease of wheat. Identification of novel stripe-rust resistance genes and cultivation of resistant varieties are considered to be the most effective approaches to control this disease. In this study, we evaluated the infection type (IT), disease severity (DS) and area under the disease progress curve (AUDPC) of 143 Chinese wheat landrace accessions for stripe-rust resistance. Assessments were undertaken in five environments at the adult-plant stage with Pst mixture races under field conditions, in addition, IT was assessed at the seedling stage with two prevalent Pst races (CYR32 and CYR34) under controlled greenhouse conditions. Results:Seventeen accessions showed stable high-level resistance to stripe rust across all environments under field tests,while four accessions showed resistance to the Pst races CYR32 and CYR34 at the all-stage.Combining phenotypic data from the field and greenhouse trials with 6404 markers covering the whole genome, we detected 17quantitative trait locus(QTLs) on 11 chromosomes for IT associated with seedling resistance and 15 QTLs on seven chromosomes for IT, final disease severityor AUDPC associated with adult-plant resistance. Four stable QTLs detected on four chromosomes, which explained 9.99%–23.30% of the phenotypic variation, were simultaneously associated with seedling and adult-plant resistance. Integrating a linkage map of stripe-rust resistance in wheat, 29 QTLs overlapped with previously reported genes or QTLs, while two and oneQTLs conferring seedling and adult-plant resistancerespectively were mapped distantly from previously reported stripe rust resistance genes or QTLs and may be novel resistantloci. Conclusions:Our results provided an integrated view of stripe rust resistance resources in a Chinese wheat landrace diversity panel from the southern autumn-sown spring wheat zone.The identified resistant accessions and resistance loci will be useful in the ongoing effort to develop new wheat cultivars with strong resistance to stripe rust.
