BackgroundHydrogen gas (H 2 ) and strigolactones (SLs) are involved in various biotic and abiotic stress response in plants. However, the crosstalk between H 2 and SLs has not been investigated. MethodsUsing pharmacological methods and virus-induced gene-silencing, the regulatory roles of H 2 and SLs and their interaction in tomato (Solanum lycopersicum L. 'Micro-Tom') under salt stress were investigated. ResultsBoth GR24 (a SLs synthetic analog) and hydrogen rich water (HRW, a H 2 donor) signi cantly reversed salt-induced growth retardation as evidenced by promoted root morphological parameters and root activity. SLs might be involved in H 2 -enhanced salt stress tolerance in tomato seedling roots. Additionally, HRW treatment increased endogenous SLs content in tomato seedling roots under salt stress However, the positive roles of HRW were blocked by TIS108 (a speci c SLs synthesis). In addition, HRW and GR24 could effectively maintain the integrity of the internal anatomical structure in roots under salt stress; while TIS108 also inhibited the positive roles of HRW. Thus,. Simultaneously, HRW treatment signi cantly up-regulated the expression levels of SL biosynthesis-related genes SlCCD7, SlCCD8, SlD27 and SlMAX1 and SL signal transduction genes SlD14 and SlMAX2 under salt stress. Further, after silencing SlD27 and SlMAX1 genes, the alleviation effect of HRW on tomato roots under salt stress was basically eliminated. HRW did not increase the content of endogenous SLs in SlD27 and SlMAX1 silenced seedlings. ConclusionSLs biosynthesis genes SlMAX1 and SlD27 may be involved in H 2 -alleviated salt stress in tomato seedlings.

SUMMARYPoor management and excess fertilization of apple (Malus domestica Borkh.) orchards are causing increasingly serious soil acidification, resulting in Al toxicity and direct poisoning of roots. Strigolactones (SLs) are reported to be involved in plant responses to abiotic stress, but their role and mechanism under AlCl3 stress remain unknown. Here, we found that applying 1 μm GR24 (an SL analoge) significantly alleviated AlCl3 stress of M26 apple rootstock, mainly by blocking the movement of Al through cell wall and by vacuolar compartmentalization of Al. RNA‐seq analysis identified the core transcription factor gene MdWRKY53, and overexpressing MdWRKY53 enhanced AlCl3 tolerance in transgenic apple plants through the same mechanism as GR24. Subsequently, we identified MdPMEI45 (encoding pectin methylesterase inhibitor) and MdALS3 (encoding an Al transporter) as downstream target genes of MdWRKY53 using chromatin immunoprecipitation followed by sequencing (ChIP‐seq). GR24 enhanced the interaction between MdWRKY53 and the transcription factor MdTCP15, further increasing the binding of MdWRKY53 to the MdPMEI45 promoter and inducing MdPMEI45 expression to prevent Al from crossing cell wall. MdWRKY53 also bound to the promoter of MdALS3 and enhanced its transcription to compartmentalize Al in vacuoles under AlCl3 stress. We therefore identified two modules involved in alleviating AlCl3 stress in woody plant apple: the SL‐WRKY+TCP‐PMEI module required for excluding external Al by blocking the entry of Al3+ into cells and the SL‐WRKY‐ALS module allowing internal detoxification of Al through vacuolar compartmentalization. These findings lay a foundation for the practical application of SLs in agriculture.

Cotton fiber, a crucial and sustainable resource for global textile production, undergoes a complex five-stage developmental process, encompassing initiation, elongation, transition, secondary cell wall biosynthesis, and maturation. These elongated single-cell fibers originate from the outer ovule epidermis. The development of cotton fibers involves intricate changes in gene expression and physiological processes, resulting in a nearly pure cellulose product vital for the global cotton industry. Decoding genes associated with fiber development enhances our understanding of cotton fiber mechanisms and facilitates the cultivation of varieties with enhanced quality. In recent decades, advanced omics approaches, including genomics, transcriptomics, and proteomics, have played a pivotal role in identifying genes and gene products linked to cotton fiber development, including the MYB transcription factor family which coordinates cotton fiber development. Molecular studies have revealed transcription factors like MYB, WRKY, Homeodomain Leucine Zipper (HD-ZIP) and basic Helix-Loop-Helix (bHLH) influencing fiber initiation and elongation. The intricate interplay of phytohormones like auxin, gibberellic acid (GA), brassinosteroid (BR), jasmonic acid (JA), ethylene, abscisic acid (ABA), and cytokinin is explored, providing a comprehensive perspective on shaping cotton fibers. Numerous candidate genes and cellular processes affecting various aspects of fiber development hold promise for genetic engineering or marker-assisted breeding to improve fiber quality. This review presents a comprehensive overview of key achievements in cotton molecular biology, with a specific emphasis on recent advancements in understanding transcription factors and phytohormones involved in cotton fiber initiation and elongation.
