BACKGROUND: Chronic hyperglycemic conditions can activate aberrant metabolic pathways causing neurotoxicity. OBJECTIVE: This study aimed to explore the effect of ethanol extract of Hibiscus sabdariffa calyxes (HS) against high glucose-induced neurotoxicity in Neuro-2a cells and Caenorhabditis elegans. METHODS: To ascertain the neuroprotective effect, Neuro-2a cells were pre-treated with HS followed by high glucose and assessed for cell viability, reactive oxygen species (ROS) generation, alterations in mitochondrial membrane potential (ΔΨm) using confocal microscopy, Real-Time PCR, Western blot and in silico approaches for the compounds identified through LC-MS/MS analysis. Further, C. elegans were treated with HS extract in the presence of glucose and analyzed for the neuroprotective effect. RESULTS: High glucose exhibited toxicity in Neuro-2a cells by ROS generation, disrupting ΔΨm, modulating stress response and lipid metabolism genes, altering signaling proteins (AKT, JNK), and apoptosis (P53, Caspase-3). However, pre-treatment with HS extract reversed the effect and exhibited neuroprotection. Compounds including allo-Aromadendrene, and N-Feruloyltyramine were identified through LC-MS/MS analysis. Docking studies against candidate protein targets indicated that the compounds of HS extract exhibit higher docking scores and can inhibit/activate the targets. Further, HS extended the lifespan of C. elegans (CL2006) from high glucose toxicity through the downregulation of A β. CONCLUSION: Our results propose that HS with its active constituents can be considered a promising therapeutic agent to treat hyperglycemia associated neurodegenerative diseases.

Background: Neurite outgrowth is an important process in neural reorganization and repair after neuronal injury. Neurite outgrowth is one of the important mechanisms to maintain normal physiological neuronal function. Neurite stimulation may help to prevent or rehabilitate brain regions in neurodegenerative disease. Objectives: The aim of this study was to screen selected ethnopharmacological herbs for stimulatory effects on neurite outgrowth and to test for any cytotoxicity and phytochemical properties. Materials and methods: The herbal extracts derived from Acanthus ebracteatus Vahl. leaves, Carthamus tinctorius L. flower, and Streblus asper Lour. bark was tested for neurite outgrowth stimulation/potentiation and cytotoxic and phytochemical properties. Results: The extract of Carthamus tinctorius L. flowers at concentrations of 50 and 500 µg/mL could significantly stimulate potentiation of neurite outgrowth in Neuro-2a cells whereas other extracts could not. We found that treatment of the cells with a concentration up to 500 µg/mL of the Carthamus tinctorius L. extract showed no cytotoxicity. Conclusion: The neurite potentiation effect might be due to other chemical constituents rather than phytochemical properties, especially total flavonoid, and phenolic contents, and antioxidant activity of the Carthamus tinctorius L. extract. The result showed that Carthamus tinctorius L. flowers extract could be a good candidate for use as a drug protecting against neuronal damage and neurodegenerative disease since it provides low cytotoxicity and neurogenic enhancement.

Alzheimer's disease (AD) is one of the progressive neurodegenerative disorders and the most common cause of dementia in the elderly worldwide causing difficulties in the daily life of the patient. AD is characterized by the aberrant accumulation of β-amyloid plaques and tau protein-containing neurofibrillary tangles (NFTs) in the brain giving rise to neuroinflammation, oxidative stress, synaptic failure, and eventual neuronal cell death. The total cost of care in AD treatment and related health care activities is enormous and pharmaceutical drugs approved by Food and Drug Administration have not manifested sufficient efficacy in protection and therapy. In recent years, there are growing studies that contribute a fundamental understanding to AD pathogenesis, ADassociated risk factors, and pharmacological intervention. However, greater molecular process-oriented research in company with suitable experimental models is still of the essence to enhance the prospects for AD therapy and cell lines as a disease model are still the major part of this milestone. In this review, we provide an insight into molecular mechanisms, particularly the recent concept in gut-brain axis, vascular dysfunction and autophagy, and current models used in the study of AD. Here, we emphasized the importance of therapeutic strategy targeting multiple mechanisms together with utilizing appropriate models for the discovery of novel effective AD therapy.
