Stratum corneum lipids liposomes for the topical delivery of 5-aminolevulinic acid in photodynamic therapy of skin cancer: preparation and in vitro permeation study

Abstract: BackgroundPhotodynamic therapy (PDT) using 5-aminolevulinic acid (5-ALA) is a skin cancer therapy that still has limitations due to the low penetration of this drug into the skin. We have proposed in this work a delivery system for 5-ALA based on liposomes having lipid composition similar to the mammalian stratum corneum (SCLLs) in order to optimize its skin delivery in Photodynamic Therapy (PDT) of skin cancers.MethodsSCLLs were obtained by reverse phase evaporation technique and size distribution of the vesi… Show more

“…Accumulation of ROS in substantial concentrations damages cellular DNA and/or membranes, and induces cell death. [9][10][11][12][13][14] We have previously reported that ALA-PDT could effectively inhibit HuCC-T1 cell survival via increased production of intracellular ROS. 19 To further improve the effect of ALA-PDT on HuCC-T1 cells, we sought to boost ROS levels after PDT.…”

“…[3][4][5][6]9,10 Excitation of a photosensitizer by irradiation with a suitable light source can produce a significant amount of reactive oxygen species (ROS). 9,10 ROS elicit DNA and cell membrane damage through lipid peroxidation/alterations in membrane fluidity. The damage to cellular organelles, such as mitochondria and microsomes, can be harnessed to increase the death of tumor cells.…”

“…This fact reduces or avoids the risk of prolonged phototoxicity to the human body. [14][15][16][17] Recently, researchers reported the effects of ALA-based PDT in various tumor cells such as oral squamous cell carcinoma, melanoma, glioma, lymphoma, esophageal carcinoma, and basal cell carcinoma.…”

“…Essa barreira é formada pelos corneócitos que são células inativas formadas no estágio final de diferenciação das células da epiderme . Desta forma, esta camada não é o alvo para fármacos tópicos que têm ação em receptores localizados na epiderme viável e derme, como é o caso do ácido retinóico (PIERRE et al, 2001;). …”
