Stratum basale keratinocyte expression of the cell-surface glycoprotein CDCP1 during epidermogenesis and its role in keratinocyte migration

Abstract: These findings delineate the expression of CDCP1 in human epidermal keratinocytes during epidermogenesis and demonstrate that CDCP1 is involved in keratinocyte migration.

“…Incubation at 95 C for 4 minutes in 10 mmol/L Sodium Citrate buffer (pH 6.0; NuMA and vWF), or at 98 C for 20 minutes in Tris-EDTA buffer (pH 9.0; VN and IGFBP-3), were used to unmask the antigens. Immunohistochemistry was performed using the MACH 4 Universal Detection System (Biocare Medical) as described previously (21). Briefly, primary antibodies against NuMA (1:200), vWF (1:400), VN (1:300), and IGFBP-3 (1:150) were diluted in Da Vinci Green Diluent (Biocare Medical), and incubated overnight on the sections at 4 C. Positive immunoreactivity was visualized using the 3,3-diaminobenzidine (DAB; Biocare Medical) substrate, resulting in formation of a brown pigment.…”

“…In vivo, basal epidermal keratinocytes are responsible for natural wound healing . Interestingly, immunostainings of human skin sections showed an exclusive expression of OR51B5 in suprabasal keratinocytes, whereas OR2A4/7 is expressed in suprabasal keratinocytes and basal melanocytes.…”

“…Formalin‐fixed, paraffin‐embedded sections (5 μm) of both human skin and HSE models were processed for IHC. Heat‐induced epitope retrieval was performed in a Decloaking Chamber ™ (Biocare Medical, Concord, CA, U.S.A.) and IHC was performed using the MACH 4 ™ Universal Detection System (Biocare Medical), as described previously . Primary antibodies against keratin 1 (1 : 500), loricrin (1 : 1000), Ki‐67 (1 : 100), KLK7 (1 : 400), DSC1 (1 : 1000), CDSN (1 : 1000), LEKTI (1 : 500) and SKALP (1 : 500) were diluted in Da Vinci Green Diluent ™ (Biocare Medical) and applied to the sections for 60 min at 37 °C or overnight at 4 °C.…”
