Strategies in relative and absolute quantitative mass spectrometry based proteomics

Abstract: Quantitative mass spectrometry approaches are used for absolute and relative quantification in global proteome studies. To date, relative and absolute quantification techniques are available that differ in quantification accuracy, proteome coverage, complexity and robustness. This review focuses on most common relative or absolute quantification strategies exemplified by three experimental studies. A label-free relative quantification approach was performed for the investigation of the membrane proteome of sen… Show more

“…A range of LC‐MS‐coupled techniques have been developed to perform rapid, robust, large‐scale, and high‐throughput analyses . Indeed, they are well known to address questions such as protein–protein interactions (PPI), protein abundances, or post‐translational modifications between two or more physiological states (e.g., healthy/disease) . Shotgun proteomics have become a powerful analytical technique for protein identification in complex biological samples.…”

“…To date, sample preparation is managed without labeling reagents, which means it is a simple and cost‐efficient technique. Relative label‐free methods can roughly be subdivided into counting the number of peptide‐to‐spectrum matches (PSMs; spectral counting) obtained for each protein, as more abundant proteins are more likely to be observed in peptide spectra, or measuring peptide signal intensities by using the extracted ion chromatogram (XIC) . The main advantage of label‐free quantification is the possibility to measure and compare an unlimited number of samples and to analyze untreated proteins or peptides with a low amount of sample.…”

“…Relative quantification compares relative protein abundance between different biological conditions (see reviews in refs. []). Quantitative proteomic analyses coupled to MS are based on stable isotope labelling strategies or label free quantification (LFQ).…”

“…[30] Mit hçherem Druck und der Verwendung von Elektrospray-Ionisation werden nun zehntausende Peptide in einem 60-bis 120-minütigen Lauf mit einer Vielzahl von Spektren identifiziert, die fürd ie Identifizierung eines Peptids bençtigt werden. Aufgrund des Bedarfs in Biologie und Medizin wurden daraufhin Totvolumen und Tr ennsäulen verkleinert.…”
