Strategies for directing the structure and function of three-dimensional collagen biomaterials across length scales

Abstract: Collagen type I is a widely used natural biomaterial that has found utility in a variety of biological and medical applications. Its well characterized structure and role as an extracellular matrix protein make it a highly relevant material for controlling cell function and mimicking tissue properties. Collagen type I is abundant in a number of tissues, and can be isolated as a purified protein. This review focuses on hydrogel biomaterials made by reconstituting collagen type I from a solubilized form, with an… Show more

“…Collagen is often used as an excellent matrix for cell attachment due to its integrin binding sites [89]. The threedimensional porous structure of collagen hydrogels allows sufficient exchange of oxygen, wastes and nutrients inside the scaffold [90] for short-term or long-term MCS culture [91].…”

“…12,38 Acid solubilization is most commonly used for minimally crosslinked collagens, such as rat tail tendon, while a combination of neutral salt solution with proteolytic extraction (pepsin digestion) is often used in order to fully denature highly crosslinked collagens from bovine or porcine skin. 11,20,39 Many combinations of collagen sources, solubilization techniques, and solvents are in use for preparation of collagen stock solutions. Further, collagen stock solutions, even those produced commercially, have been found to have significant batch-to-batch variation.…”

“…A quitosana, é um exemplo desse tipo de material, sendo muito utilizado para a fabricação de scaffolds na reparação de tecidos [1,4,5]. A quitosana é um copolímero composto por N-acetil-2-amino-2-desoxi-D-glucopiranose e 2-amino-2-desoxi-D-glucopiranose, onde os dois tipos de unidades de repetição estão ligadas por (1→4)-β-glicosídica, estas unidades estão distribuídas em diferentes graus, dependendo das porções acetiladas.…”
