Strain-specific and tissue-specific expression of mouse mast cell secretory granule proteases.

Abstract: As assessed by RNA blot analyses with genespecific probes, we report that the perivascular connective tissue mast ceOls (CTMCs) in the ear and skin of BALB/cJ mice contain abundant levels of the mouse mast cell protease (mMCP) 7 transcript, in addition to those protease transcripts present in their serosal mast cells (SMCs). High levels of the mMCP-7 transcript also were detected in the ears of WBB6F1/J-+/+, WCB6F1/J-+/+, WB/ReJ-+/+, and WC/ReJ-+/+ mice. However, the ears of these four strains and the SMCs fro… Show more

“…Molecules other than Ubc9 would be responsible for MITF-induced degradation of Smads. mMCP-7 is expressed predominantly in a subpopulation of differentiated mast cells that reside in numerous connective tissue sites (4,57,58). mMCP-7 is a tryptase that degrades fibrinogen (59).…”

“…Preliminary studies confirmed that MC-deficient W/W v mice possess a greatly diminished ability to kill KP1415 bacteria relative to MC-sufficient WBB6F1 ϩ/ϩ mice or C57BL/6 mice (data not shown). It has been shown previously that the MCs in WBB6F1 ϩ/ϩ and C57BL/6 mice differ in their expression of mMCP-7, but not of mMCP-6 (47)(48)(49). The finding that WBB6F1 ϩ/ϩ and C57BL/6 mice do not differ significantly in their ability to combat a KP1415 bacterial infection now indicates that the primary immunologic defect in MC-deficient W/W v mice is not a consequence of their failure to express mMCP-7.…”

“…These three MC tryptases also can be differentially expressed within specific tissues of the same mouse strain. For example, the safranin ϩ MCs in the skin of BALB/cJ mice contain abundant amounts of mMCP-6 and mMCP-7, whereas the safranin ϩ MCs in the peritoneal cavity of this mouse strain express mMCP-6 but not mMCP-7 (29). The intestines of C57BL/6J mice also contain more Prss31 than mMCP-6 or mMCP-7 (15).…”
