Stimulation with the Aureobasidium pullulans-produced β-glucan effectively induces interferon stimulated genes in macrophage-like cell lines

Abstract: A β-(1,3),(1,6)-D-glucan produced by A. pullulans (AP-PG) is known to be an immune stimulating agent. In this study, we demonstrate that the stimulation with AP-PG effectively induces the interferon (IFN) stimulated genes (ISGs) in macrophage-like cell lines. The ISGs, Mx1, ISG15, and viperin mRNAs were significantly increased in RAW264.7 cells after stimulation with AP-PG. The stimulation with AP-PG transiently induced IFN-β mRNA. However, the expression of viperin mRNA was also increased after stimulation wi… Show more

“…These pathways consist of the activation of transcription factors, such as nuclear factor kappa-light-chain-enhancer (NF-κB) of activated B cells and interferon regulatory factors (IRFs), which results in production of type I IFNs and pro-inflammatory cytokines. For instance, a type I IFN gene signature was promoted in human DCs that were stimulated with WGP and curdlan [69] as well as in human and murine macrophage cell lines stimulated with a β-glucan derived from A. pullulan [70]. More specifically, the intracellular receptors NOD1 and NOD2 can interact with the inflammasome NLRP3, which then leads to production of IL-1 cytokines [71].…”

“…The A. pullulans incubation fluid containing β-glucan as the main ingredient is consumed as a supplementary food in many countries as it represents the activation of the immune-stimulation. Also, A. pullulans incubation fluid is regarded as useful and effective for delaying the outbreaks of many diseases, and has been reported to show the activation of anti-tumor (Kimura et al, 2006), anti-allergy (Kimura et al, 2007), and anti-inflammation (Muramatsu et al, 2014) in the mouse model.…”

“…As noted above, the antiviral effects of APβG may also be realized through the expression of interferon-stimulated genes (ISGs) in macrophage-like cell lines. These findings suggest that APβG stimulation effectively promotes the expression of ISGs through inducing IFNs and enhancing STAT1-mediated transcriptional activity [ 50 ].…”
