Stimulation of Inducible Nitric Oxide by Hepatitis B Virus Transactivator Protein HBx Requires MTA1 Coregulator

Abstract: Nitric oxide has been implicated in the pathogenesis of inflammatory disorders, including hepatitis B virus-associated hepatocellular carcinoma. Transactivator protein HBx, a major regulator of cellular responses of hepatitis B virus, is known to induce the expression of MTA1 (metastasis-associated protein 1) coregulator via NF-B signaling in hepatic cells. However, the underlying mechanism of HBx regulation of the inducible nitric-oxide synthase (iNOS) pathway remains unknown. Here we provide evidence that MT… Show more

“…HBx has been shown to induce the expression of metastasis-associated protein 1 (MTA1) coregulator, which is a master chromatin modifier involved in carcinogenesis and a positive regulator of inducible nitric oxide synthase (iNOS) transcription [57,80]. The HBx/MTA1 complex stimulates the production of NO, which has been implicated in hepatocarcinogenesis [57]. HBx has also been demonstrated to increase the expression of cyclooxygenase-2 (COX-2) mRNA and protein [81].…”

“…Secondly, the upregulation of inflammatory mediators by HBx is another pathway of interest. The HBx/MTA1 complex stimulates the production of NO, which has been implicated in hepatocarcinogenesis [57].…”

“…It is a component of the nuclear remodeling and histone deacetylase complex involved in carcinogenesis. MTA1 is a positive regulator of inducible nitric oxide synthase (iNOS) transcription as well (Bui-Nguyen et al, 2010). The HBx/MTA1 complex stimulates the production of nitric oxide (NO°), by stimulation of iNOS expression in an NF-κB-dependent manner.…”

“…Several reports represented a positive correlation between the expression of NF-B and TG2 expression (32,33) and between the expression of NF-B and MTA1 (8,16,34), suggesting the existence of a positive regulatory mechanism among these genes. Increased TG2 activity triggers NF-B activation by inducing the polymerization of I-B␣ rather than stimulating I-B␣ kinase (35).…”
