Stimulation of Glycosphingolipid Biosynthesis by L-Threo-1-Phenyl-2-Decanoylamino-1-Propanol and Its Homologs in B16 Melanoma Cells

Abstract: Previous studies have demonstrated that the ceramide analog D-threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (D-threo-PDMP) inhibits glucosylceramide (GlcCer) synthase and thus leads to extensive depletion of glycosphingolipids (GSLs) biosynthesized from GlcCer [reviewed by Radin, N.S., Shayman, J.A., and Inokuchi, J. (1993) Adv. Lipid Res. 26, 183-213). In the present study, stereospecificity of PDMP activity was demonstrated with an enantiomeric pair, D-threo-PDMP and L-threo-PDMP. Treatment of B16 m… Show more

“…L-threo-PDMP, an isomer of D-threo-PDMP, was also studied. It stimulated ganglioside synthesis, increasing cellular ganglioside content from 3.95 Ϯ 0.39 to 5.06 Ϯ 0.64 (n ϭ 3) nmol of LBSA/mg of protein when the cells were exposed to 20 M L-PDMP for 6 days, which is consistent with a previous report (23). However, it had no effect on neurite formation, nor did it inhibit retinoic acidinduced neurite formation (Fig.…”

“…D -PDMP was initially suggested to be a selective inhibitor of GlcT-1 with an IC 50 of ∼10 µ M [23], but subsequent studies revealed that it also inhibits GalT-2 (with equal potency), two of the globo-series synthases [15], and the sialyltransferase that converts LacCer to ganglioside GM3 [24]. Treatment of NIH 3T3 fibroblasts or kidney proximal tubular cells with D -PDMP reduced the cellular levels of GlcCer and LacCer and increased the levels of Cer in a concentration- and time-dependent fashion [15, 25].…”

“…H4APPsw cells were treated with L-PDMP, an activator of GCS (Inokuchi et al, 1995(Inokuchi et al, , 1997 in order to assess whether the inhibition of GSK3β activity observed in D-PDMP-treated cells might be linked to reduced cellular ganglioside levels. We first assessed that the enantiomer L-PDMP properly stimulated ganglioside synthesis in our experimental settings (Fig.…”
