Stimulation of Bone Healing by Sustained Bone Morphogenetic Protein 2 (BMP-2) Delivery

Abstract: The aim of the study was to investigate the effect of a sustained release of bone morphogenetic protein2 (BMP-2) incorporated in a polymeric implant coating on bone healing. In vitro analysis revealed a sustained, but incomplete BMP-2 release until Day 42. For the in vivo study, the rat tibia osteotomy was stabilized either with control or BMP-2 coated wires, and the healing progress was followed by micro computed tomography (μCT), biomechanical testing and histology at Days 10, 28, 42 and 84. MicroCT showed a… Show more

“…It can induce new bone formation in vivo, and have a strong osteoinductive effect on human bone marrow stromal cells in vitro . Therefore, researches on BMP‐2 would be helpful for understanding bone formation, fracture healing, and bone growth around implants . However, BMP‐2 has a very short half‐life and is easy to degenerate after implantation in the human body .…”

“…The most utilized drug in bone tissue engineering studies has been by far bone morphogenetic protein (BMP). In particular, BMP-2 is known to play an essential role in bone healing, namely by influencing osteogenesis as well as vascularization [3]. This effect has been clearly demonstrated in various studies where BMP-2 was loaded into various types of implantable carriers [4, 5•, 6, 7].…”

“…In different studies, vancomycin was incorporated into polyurethane or chitosan materials for local delivery to treat and prevent infection (Li, Brown, Wenke, & Guelcher, ; Stinner, Noel, Haggard, Watson, & Wenke, ), whereas another group combined vancomycin delivered in a fibrin gel with alginate beads to deliver stem cells (Hou, Xu, Li, Feng, & Zen, ). In addition to antibiotics, growth factors such as bone morphogenetic proteins are often included in bone regenerative strategies for osteoinduction (Brown et al, ; Faßbender, Minkwitz, Strobel, Schmidmaier, & Wildemann, ; Jiang et al, ; Li, Vepari, Jin, Kim, & Kaplan, ). The efficacy of these complex systems however must be confirmed at the scaffold level due to the effects of infection on the graft itself.…”
