Steroid Transport, Local Synthesis, and Signaling within the Brain: Roles in Neurogenesis, Neuroprotection, and Sexual Behaviors

Abstract: Sex steroid hormones are synthesized from cholesterol and exert pleiotropic effects notably in the central nervous system. Pioneering studies from Baulieu and colleagues have suggested that steroids are also locally-synthesized in the brain. Such steroids, called neurosteroids, can rapidly modulate neuronal excitability and functions, brain plasticity, and behavior. Accumulating data obtained on a wide variety of species demonstrate that neurosteroidogenesis is an evolutionary conserved feature across fish, bi… Show more

“…As cholesterol is an important component of many cellular membranes and is also the substrate for the synthesis of bile acids, steroid hormones and regulating oxysterols, disruption of cholesterol transport may have widespread consequences in cholesterol homeostasis (Diotel et al, 2018;McDonald & Russell, 2010;McMillan & DeMorrow, 2016).…”

“…Sex hormones are critical modulators of adult neuroplasticity and behavior. Sex hormones can either be produced locally in the brain or be synthesized in the gonads and reach the brain through blood circulation (Diotel et al, 2018). Additionally, circulating testosterone can be converted in the brain into its active metabolites through enzymatic action: into DHT (5αdihydrotestosterone) by 5α-reductase, or into estradiol (17β-estradiol) via the enzyme aromatase.…”

Through their interactions with androgen and estrogen receptors in the brain, sex hormones can mold brain circuits, and thus shape animal behavior in complex ways (Chen et al, 2013;Diotel et al, 2018;Gahr, 2004;Galea, 2008).…”

“…Numerous reports suggest that estrogens are neuroprotective (Diotel et al. ) and cardioprotective (Knowlton and Lee ; Luo and Kim ) and act as anti‐oxidants that in turn sustain heart health.…”
