Steroid hormones and their respective nuclear receptors are essential mediators in numerous physiologic and pathophysiologic processes, ranging from regulation of metabolism, immune function, and reproductive processes to the development of hormone-dependent cancers such as those of the breast and prostate. Because steroids must enter cells before activating nuclear receptors, understanding the mechanisms by which cellular uptake occurs is critical, yet a clear understanding of these mechanisms has been elusive. It is generally assumed that diffusion-driven uptake is similar across various steroids whereas an elevated cellular concentration is thought to reflect active uptake, but these assumptions have not been directly tested. Here we show that intact cells rapidly accumulate free steroids to markedly elevated concentrations. This effect varies widely depending on steroid structure; more lipophilic steroids reach more elevated concentrations. Strong preferences exist for 3β-OH, Δ5-steroids vs. 3-keto, Δ4-structural features and for progestogens vs. androgens. Surprisingly, steroid-structure-specific preferences do not require cell viability, implying a passive mechanism, and occur across cells derived from multiple tissue types. Physiologic relevance is suggested by structure-specific preferences in human prostate tissue compared with serum. On the other hand, the presence of serum proteins in vitro blocks much, but not all, of the passive accumulation, while still permitting a substantial amount of active accumulation for certain steroids. Our findings suggest that both passive and active uptake mechanisms make important contributions to the cellular steroid uptake process. The role of passive, lipophilicity-driven accumulation has previously been largely unappreciated, and its existence provides important context to studies on steroid transport and action both in vitro and in vivo.

The small size of microplastics and their wide distribution in water environments have attracted worldwide attention and heated discussion, because of their ingestion by aquatic organisms. At present, there are few studies on microplastics pollution in freshwater aquaculture ponds, especially shrimp ponds. In this study, the aquaculture ponds in the Pearl River Estuary were investigated. The abundance and composition of microplastics in different environmental media were studied to explore the potential sources and risk levels of microplastics, so as to provide basic data for the study of microplastics pollution in aquaculture ponds. Microplastics were observed in water and sediment samples at all sampling sites, with the abundance of 6.6 × 103–263.6 × 103 items/m3 (surface water) and 566.67–2500 items/kg (sediment), respectively. Thirty-seven individuals collected in six ponds belong to four species. Microplastics were observed in the gastrointestinal tract (GIT) of all fishes and shrimps, with the abundance ranging from 3–92 items/individual (fish) and 4–21 items/individual (shrimp). Among all samples, microplastics with the size range of <1 mm and fiber shape were the most common. The main microplastic components were cellulose, polypropylene (PP), and polyethylene (PE). The results of potential risk assessment showed that the pollution investigation of microplastics should not only consider the abundance. Low abundance does not mean low risk. Taking the toxicity score and abundance of microplastics as evaluation indexes to reflect the pollution status of microplastics may make the results more reliable.

Steroid hormones in the aquatic environment may pose a risk to fish health due to their ubiquitous presence and high biological activity. At present, the uptake process and toxicokinetics of steroids in fish are poorly known, in particular the role of cell membrane transporters. We investigated the interaction of 17 endogenous and environmental steroids with the zebrafish organic anion transporting peptide (Oatp1d1) uptake transporter, which is prominently expressed in liver and kidneys. We selected steroids of different classes including androstenedione (A4), progesterone (P4), and its metabolites, as well as glucocorticoids and spironolactone, and analyzed their interaction with Oatp1d1 by competitive inhibition of the uptake of the fluorescent substrate Lucifer Yellow. The half-maximal inhibition (IC50) values derived from sigmoid inhibition curves were lowest for P4, and the order of increasing IC50 values was as follows: 17α-hydroxyprogesterone > clobetasol propionate > spironolactone > 21α-hydroxyprogesterone > fludrocortisone acetate and additional glucocorticoids. The interaction activity showed a positive correlation with the lipophilicity of the steroids. Our data show that different classes of steroids interact with Oatp1d1 with different activity (either by uptake or inhibition, or both). This is of importance, because in consequence, steroids may interfere with the transport of endogenous substrates, and thus physiological processes. Moreover, steroids may alter cellular trafficking of environmental contaminants by competitive inhibition of this transporter. Environ Toxicol Chem 2018;37:2670-2676. © 2018 SETAC.
