Stem cells for organoids

Abstract: Organoids are three‐dimensional (3D) cell culture systems that simulate the structures and functions of organs, involving applications in disease modeling, drug screening, and cellular developmental biology. The material matrix in organoids can provide a 3D environment for stem cells to differentiate into different cell types and continuously self‐renew, thereby realizing the in vitro culture of organs, which has received extensive attention in recent years. However, some challenges still exist in organoids, i… Show more

“…Immunohistochemistry: According to the method reported previously, the immunohistochemistry of the proximal left tibias after decalcification and the colons of rats were performed and assessed. [51,56,57] In brief, the sections were incubated with NLRP3 (1:200, Servicebio, China), Caspase-1 (1:300, Servicebio, China), IL-1𝛽 (1:100, Servicebio, China), IL-18 (1:200, Servicebio, China), Occludin (1: 600, Servicebio, China), and ZO-1 (1: 600, Servicebio, China), respectively. All images were captured by a LSCM (Zeiss, Germany).…”

“…Nowadays, engineered microbes have served as significant therapeutic agents for various therapeutic applications [113][114][115][116][117]. Several internal and external stimuli can activate the colonization and payload release of these administered bacteria [118,119].…”

“…Not only that, but iPSC can also avoid the ethical problems of ESC extraction from patients. [86] In the last decade or so, with the breakthrough discovery of iPSCs, researchers have differentiated them into a variety of cells with different functions and phenotypes. iPSC gradually showed great potential for applications in the direction of drug testing, disease modeling, and regenerative medicine [87,88] (Figure 7D, E).…”

“…[104] In contrast to iPSC, ASC-derived organoids are mainly composed of mature cells, which tend to maintain high genetic stability after long-term culture. [86] However, the differentiation potential of ASC is inferior to that of iPSC, and the derived organoids are often only transformed into a single epithelial cell type. Boonekamp et al [105] used epithelial stem cells, determined optimal culture conditions, and successfully established a human epidermal organoid model (Figure 10Ci) with the promotion of cytokines such as fibroblast growth factor (FGF).…”
