Stem Cell-Based Therapies for Inflammatory Bowel Disease

Abstract: Inflammatory bowel disease (IBD) is a chronic, relapsing disease that severely affects patients’ quality of life. The exact cause of IBD is uncertain, but current studies suggest that abnormal activation of the immune system, genetic susceptibility, and altered intestinal flora due to mucosal barrier defects may play an essential role in the pathogenesis of IBD. Unfortunately, IBD is currently difficult to be wholly cured. Thus, more treatment options are needed for different patients. Stem cell therapy, mainl… Show more

“…The utility of stem cell technology for the repair and regeneration of the injured intestine has been a major focus, and stem cell-based regenerative therapies hold promise for anastomotic healing (Flatres et al, 2019;Lightner, 2019;Kent et al, 2021;Zhang et al, 2022). In fact, studies have reported the utilization of stem cell patches in animal experiments to promote anastomotic healing with positive outcomes (Maruya et al, 2017;Van de Putte et al, 2017;Sukho et al, 2018;Morgan et al, 2020;Pan et al, 2020).…”

“…Stem cell-based regenerative therapies are considered a promising scheme as stem cells have the ability to differentiate into intestinal cells or promote intestinal cell proliferation (Flatres et al, 2019;Lightner, 2019;Kent et al, 2021;Zhang et al, 2022). Several preclinical studies investigated the role of stem cell transplantation in accelerating the healing of intestinal anastomosis, and preliminary results indicated that stem cell transplantation has the potential to protect surgical anastomosis (Maruya et al, 2017;Van de Putte et al, 2017;Sukho et al, 2018;Morgan et al, 2020;Pan et al, 2020).…”

“…Stem cell transplantation therapy appears to be used to improve clinical symptoms in patients with IBD when conventional therapies are ineffective, or even ineffective in the treatment of IBD. Stem cell transplantation therapy can be divided into hematopoietic stem cell transplantation (HSCT) and mesenchymal stem cell transplantation (MSCT) [ 59 ]. HSCT can be divided into autogenous and allogeneic transplantation.…”

“…Therefore, it is considered as a safer treatment [ 63 ]. Stem cell transplantation is a relatively new method of treating IBD, and more research is required to determine its safety and therapeutic effectiveness [ 59 ]. Therefore, stem cell transplantation is not yet a mainstream method used to treat IBD.…”

“…In addition to the above therapeutic mechanisms, stem cells have received more and more attention as an effective therapeutic tool with intrinsic repair ability toward IBD treatment either with or without nanomaterial loads. [ 147 ] Plenty of studies have shown that different types of stems cells including hematopoietic stem cells (HSC), mesenchymal stem cells (MSC), and intestinal epithelial stem cells can improve intestinal lesions by immunomodulation and tissue repair after homing to the inflamed intestine. Despite stem cell‐related IBD therapy still being in its fancy, it has shown huge potential in alleviating mucosal inflammation, which results in the clinical remission of IBD.…”
