Stem cell and lung cancer development: blaming the Wnt, Hh and Notch signalling pathway

Abstract: Primary lung cancer may arise from the central (bronchial) or peripheral (bronchiolo-alveolar) compartments. However the origins of the different histological types of primary lung cancer are not well understood. Stem cells are believed to be crucial players in tumour development and there is much interest in identifying those compartments that harbour stem cells involved in lung cancer. Although the role of stem cells in carcinogenesis is not well characterised, emerging evidence is providing new insights int… Show more

“…It is also involved in maintenance of normal skin structure and function, through control of the permeability barrier function (Demehri et al, 2009a; Demehri et al, 2009b; Dumortier et al, 2010). This pathway appears to play an equally important regulatory role in the oral (Papagerakis et al, 2014), esophageal (Croagh et al, 2014) and bronchial epithelia (Garcia Campelo et al, 2011). …”

“…Wnt1 and Wnt2 are considered proto-oncogenes but are also critical for cell differentiation and development during embryogenesis. In contrast, investigators have discovered that Wnt-7a, a possible tumor suppressor, is downregulated in these same cells, providing the growth advantage that is a hallmark of cancer cells [9,66,67,68]. …”

“…6 Present study indicates that the removal of CSCs is required to completely eliminate a tumor. 7 Additionally, high CSC expression of ATP-binding cassette, subfamily G member 2 (ABCG2) 8 and ATP-binding cassette, subfamily B, member 1 (ABCB1), 9 which can efflux chemotherapy drugs, are associated with chemoresistance, while activation of the Notch pathway 10 and Wnt/beta-catenin pathway 11 are linked to radioresistance in breast cancer. Characterization of the biological mechanisms leading to the development and self-renewal of BCSCs may have great clinical significance and reveal novel therapeutic targets for breast cancer.…”
