STAT4 targets KISS1 to promote the apoptosis of ovarian granulosa cells

Abstract: Background In mammals, it is known that the estradiol-17β (E2) is mainly synthetized in ovarian granulosa cells (GCs), and the excessive apoptosis of GCs induces the follicular atresia. Many studies have implicated the essential role of KISS1, with the pro-synthetic effect of E2 and the anti-apoptotic effect on GCs, in the mammalian folliculogenesis, and several STAT4 potential binding sites were previously predicted on the promoter of KISS1 in pigs. However, the biological effects of STAT4 on … Show more

“…Our study showed that STAT4 and CFLAR have positive coefficients in the model. Previous studies have shown that STAT4 plays a role in promoting apoptosis in hepatocellular carcinoma and ovarian granulosa cells ( Jiang et al, 2020 ). A study in the vascular smooth muscle cell (VSMC) showed that the effect of STAT4 on VSMC apoptosis was mainly mediated by the activation of the mitochondrial apoptotic pathway ( Lv et al, 2014 ).…”

“…GCS play an important role in oocyte development and affect female breeding performance. A number of studies have shown that small molecules including Vitamin D3 [30], FSH [31], Bisphenol A [32] and genes including CITED4 [33], CXCL12 [34], and STAT4 [35] regulate cell proliferation, apoptosis and hormone secretion in mammals, thus affecting estrus of female animals and oocyte maturation. However, as one species of the high fertility husbandry animals, the effect of CTSS gene on rabbit reproduction has been rarely studied.…”

“…After being activated, STAT4 translocates into the nucleus and regulates gene expression [33]. Previous studies have shown that STAT4 can bind to the − 305/-295 region of KISS1 promoter to negatively regulate the transcription of KISS1 in the folliculogenesis of mammals [34]. In this study, 5 putative binding sites of STAT4 on the promoter of Klotho gene were identi ed using bioinformatics analysis.…”
