Staining of mitochondrial membranes with 10-nonyl acridine orange MitoFluor Green, and MitoTracker Green is affected by mitochondrial membrane potential altering drugs

Abstract: Background: We set out to develop an assay for the simultaneous analysis of mitochondrial membrane potential and mass using the probes 10-nonyl acridine orange (NAO), MitoFluor Green (MFG), and MitoTracker Green (MTG) in HL60 cells. However, in experiments in which NAO and MFG were combined with orange emitting mitochondrial membrane potential (⌬⌿ m ) probes, we found clear responses to ⌬⌿ m altering drugs for both probes. Methods: The three probes were titrated to determine whether saturation played a role in… Show more

“…Mitochondrial turnover was assayed by staining with 10-nonyl acridine orange (NAO), a mitochondrial dye relatively insensitive to Δψ M (14,15). Staining decreased substantially after iron depletion (Fig.…”

“…Mitochondrial mass was examined with MitoTracker green FM (MTGFM; Invitrogen Canada). This probe becomes fluorescent after incorporation into the lipid environment of mitochondria, where it interacts with peptide or protein thiol groups (19). Cells were stained with 20 nM MTGFM for 45 min at 37°C in cell culture media and then analyzed by flow cytometry.…”

“…3, C (sector II) and D, Nat8l-overexpressing cells showed increased fluorescence intensity, indicating an increased mitochondrial mass. Nevertheless, others reported that MitoTracker Green changes its abilities with regard to the membrane potential (28). To ascertain that the increase in fluorescence intensity was not due to a membrane potential change, we measured membrane potential via incubation with the mitochondrial stain JC-1.…”
